Cortinarius rufo-olivaceus growth in the qinghai spruce of Helan mountain, is the joint name of any cortinariaceaes, cortinarius rufo-olivaceus is a rare edible fungus in local. At the moment, the research about cortinarius rufo-olivaceus only in analysis it’s chemical composition,but the biological activities is not reported.According to the data,a great amount of macro fungi have a obvious functions to anti-tumor in many ways. So, this research establish the H22liver cancer animal model, take the liver cancer as an example to analyze the antitumor functions of Cortinairus rufo-olivaceus and it’s possible mechanism. Also study the protection against acute liver injury of cortinarius rufo-olivaceus. The main content of the paper is as follows:1.Prepared the extraction of cortinarius rufo-olivaceus:The cortinarius rufo-olivaceus女(Pers.)Fr. was used as the material and be extracted with95%ethanol, and then system solvent extraction method is exploited, got petroleum ether, ethyl acetate and n-butanol phase; At the same time, water extraction combined with ethanol precipitation method to extract of crude polysaccharides. Finally get four parts:petroleum ether, ethyl acetate,n-butanol phase and polysaccharide.2. Antitumor activity of cortinarius rufo-olivaceus in H22tumor bearing mice:the solid tumor H22liver cancer animal model was established,find that the extraction of cortinarius rufo-olivaceus have anti-tumor activity, among that the polysaccharide part’s anti-tumor rate was more than75.24%, ethyl acetate and n-butanol phase also have high anti-tumor rate, and the tumor inhibitory effect have a same level with lentinan. Cortinarius rufo-olivaceus have no damage to immune organs, the spleen index and thymus index of mice are higher than the model control group and5-Fu group. Although5-Fu group have a highest anti-tumor rate, but show the lowest spleen index and thymus index. Detection the related protein expression by western-blot, the result shows that, cortinarius rufo-olivaceus adjusted apoptosis through changing the level of BCL-2,BAX,P53proteins, ethyl acetate phase has a obvious effect. The study also determine the cytokines in the serum of mice, such as IL-2-. TNF-α and INF-γ, ethyl acetate phase and polysaccharide group’s IL-2and INF-γ contents significantly higher than other groups(P<0.05); n-butanol phase can efficient increase the TNF-α content, up to the same level with lentinan(P<0.05).3. Study the antioxidation activities of cortinarius rufo-olivaceus in H22tumor bearing mouse: The results indicated ethyl acetate,n-butanol phase and polysaccharide part can obviously increase the activity of T-SOD,CAT,GSH-PX and decrease the concentration of MDA in liver(P<0.05);5-Fu group show the highest MDA content.4. Prevention and protection against acute liver injury by cortinarius rufo-olivaceus:The mice chemical liver injury model induced by CCL4after gavage the mice two months by the extract from cortinarius rufo-olivaceus and positive medicine control, test the related indicators in mice serum found that, cortinarius rufo-olivaceus can decrease serum ALT and AST to normal mice level, and the therapeutic effect as effective as positive control(P<0.05),especially the ethyl acetate phase and polysaccharide part. TP,ALB decreasede sharply in model group,but ethyl acetate phase and polysaccharide grpup can raise them higher than normalgroup. And cortinarius rufo-olivaceus can decrease the TBIL better than positive control and had statistical significance(P <0.05).Liver tissue antioxidant capacity show that, cortinarius rufo-olivaceus can decrease the MDA content obviously, and increase the SOD,GSH-PX up to normal mice level ((P<0.05)。 The results show that cortinarius rufo-olivaceus had positive adjustment function for immune system of organism, and control the programmed cell death regulatory,by these way to defend cancer. Cortinarius rufo-olivaceus also can enhance liver ability of oxidation and decrease lipid peroxidation level, then can protect cells from peroxidation damage and the way lead to cancer by excess free radical. |
