Effects Of Recombinant Plasmid Pegfp-afp-tk Delivered By Nano-magnetic Fluids On Targeting Therapy For Implanted Liver Cancer In Nude Mice Under Alternating Magnetic Field

Objective:under the alternating magnetic field, to study the effects of pEGFP-AFP-TK carried by PEG-PEI/Fe3O4 nano-magnetic fluid targeting therapy for liver cancer in nude mice implanted with HepG2, according the changes of tumor volume, weight, growth and the results of MVD, AFP, AI in tumor.Methods:To establish nude mice model of HepG2. Mice were randomly divided into 5 groups:the control group; the magnetic field group, nano-magnetic fluid group, pEGFP-AFP-TK group and pEGFP-AFP-TK carried by PEG-PEI/Fe3O4 nano-magnetic fluid group. Control group without any treatment; magnetic field group just under magnetic field without other treat; Different plasmids were injected into tumors according to different groups, while injecting GCV into caudal vein. At the pulse frequency of 40 kHz, and magnetic field of 5kA/m,15 minutes one day in the next 14 days. On the 7th day and the 15th day, observed the changes of tumor volume, weight growth and take pathological examination, record the results of MVD, AFP and AI in tumor, study the Thymidinekinase gene expression in heart, liver and kidney by RT-PCR.Results:The success rate to establish subcutaneous implanted liver cancer modeling nude mice is 100%. On the 7th and the 15th day, in the pEGFP-AFP-TK carried by nano-magnetic fluid group, the tumor volume, weight, serum AFP levels, and MVD expression were lower than other groups, the tumor inhibitory rate and apoptotic index significantly higher than other groups; so the pEGFP-AFP-TK carried by nano-magnetic fluid group is the best. RT-PCR measured that shows the tumor mouse heart, the lung, the kidney shows the Thymidine Kinase expression is unclear, but the tumor cell presents the Thymidine kinase expression apparently.Conclusion:Under the alternating magnetic field, pEGFP-AFP-TK carried by magnetic fluid can inhibits liver cancer remarkably. Magnetic field has certain influence to the tumor growth. What's more, magnetic field could be provided to an auxiliary treatment method for liver cancer, together with gene targeting therapy and thermotherapy for better treat results.
