Objective:To study the relationship between ApoE gene polymorphism and Yi nationality cerebral infarction in YunNan province.Compare blood lipid levels between different groups of ApoE allele types.Find regular pattern of ApoE allele frequency distribution in different nationalities at home and abroad, analysis of risk factors associated with CI incidence, to provide theoretical basis for early prevention of cerebrovascular disease.Methods:1, Choose40cases of Yi nationality cerebral infarction patients (male31cases.female9cases)in People’s hospital of Chuxiong Yi Autonomous Prefecture and People’s hospital of YaoAn country(There are30cases of atherosclerotic thrombotic cerebral infarction、8cases of lacunar cerebral infarction and2cases of artery origin cerebral embolism),and40cases of Yi nationality healthy people(male26cases,female14cases,whose age-and gender were matched with CI group) in physical examination of the People’s hospital of Chuxiong Yi Autonomous Prefecture as the objects of the study;2、Using the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method to detect and compare the ApoE genotype,and then further defined gene types by gene sequencing;3、Study the relationship between ApoE gene polymorphism and CI types;4、Study the distribution law of ApoE alleles in different nations;5、Analyze the plasma TG、TC、LDL、HDL concentration of all the research objects, compare the affect of of blood lipids by e2> ε3、ε4allele genetype.Result:1、ApoE gene polymorphism dose correlated with Yi cerebral infarction.The frequency of the ApoE4gene type (including ε3/ε4and ε4/ε4genotypes) in Yi CI patients significantly higher than those in healthy people (OR= 3.842,95%confidence interval:1.44410.219);The frequency of genotype ε3/ε3and ε3allele frequency is lower than the control group (47.5%VS77.5%,70.0%VS86.2%, P<0.05);2.ε4gene frequency (ε4(+)) associated with CI types, the s4gene frequency (ε4(+)) in atherosclerotic thrombotic cerebral infarction group is60.0%,and in lacunar cerebral infarction group is25.0%, there are differences between the two groups (P<0.05);3、The distribution of ApoE alleles frequency in different nations are different, the main distribution is closely related to geographical environment (latitude);4、TG level in CI group was obviously higher than in control group (P=0.019<0.05), but HDL-C level was obviously lower than control group (P=0.000<0.01), the TC and LDL-C have no significant difference in two groups.In all of the objects.ε2carriers have a significantly higher level of HDL-C than the ε3and ε4carriers (P<0.01), and ε4carriers have a higher level of TG and LDL than s2carriers (P<0.05).Conclusion:1. Our data show ApoE gene polymorphism is closely related to the yi nationality CI in yunnan province,and s4allele is one of the genetic susceptibility factors of CI;2、ApoE allele distribution is different in different types of CI,ε4allele is associated with atherosclerotic embolism cerebral infarction, but has no relationship with lacunar cerebral infarction;3、The ApoE e4gene frequency in crowd increased gradually with latitude increasing;4、ApoE gene polymorphisms affect blood lipid levels, subsequently lipid metabolism disorders increase the risk of atherosclerosis, and thus increase the risk of CI;5、We use both PCR-RFLP method and gene sequencing method to detect ApoE genotype, and gene sequencing method is more accurate, fast, and reliable, we can check the whole sequence by reading the bases of ApoE gene, it has the advantages of lower miss rate and good repeatability.
