Object:To investigate Apelin-13effects on HepG2cell proliferation,apoptosis, and theexpression of CyclinD1and Caspase3.This project could provide a new approch fortherapy of hepatocellular carcinoma.Method:We cultivated HepG2cells in different concentrations and duration of Apelin-13.The different concentrations of Apelin-13are①10%FBS (control),②0.0001μmol/L,③0.001μmol/L,④0.01μmol/L,⑤0.1μmol/L.And the differentdurations are①0h(control);②6h;③12h;④18h;⑤24h. Then, we detected the cellsdensity and the protein, CyclinD1and Capspase3,which is related to cell proliferationand apoptosis, by MTT and Western blot respectively.Result:1.The result of MTT shows that the Apelin-13can promote the cell proliferationwith the concentration. The OD are①0.464±0.025;②0.510±0.094;③0.547±0.174;④0.598±0.035;⑤0.654±0.016.Means are significant different.(P<0.01)2.The result of MTT shows that the Apelin-13can promote the HepG2cellproliferation with duration. In24hours,the OD are①0.489±0.030;②0.537±0.042;③0.587±0.042;④0.632±0.022;⑤0.681±0.016. Means are significantdifferent.(P<0.01)3.The result of Western blot show that Apelin-13can increase the expression ofCyclinD1with concentraton,which relative densities are①0.544±0.026;②0.699±0.015;③0.831±0.012;④0.985±0.109;⑤1.160±0.098.And the durationof Apelin-13can increase the expression of CyclinD1,that relative densities are①0.582±0.020;②0.730±0.021;③0.891±0.020;④1.059±0.064;⑤1.290±0.041. Means are significant different.(P<0.05)4.The result of Western blot show that Apelin-13can inhibit the expression of Caspase3with concetration, which relative densities are①1.024±0.092;②0.826±0.042;③0.683±0.063;④0.524±0.052;⑤0.259±0.010.And the durationof Apelin-13can increase the expression of CyclinD1,that relative densities are①0.855±0.056;②0.694±0.046;③0.446±0.020;④0.269±0.017;⑤0.189±0.009.Meansare significant different.(P<0.05)Conclusion:1.Apelin-13can induce the expression of CyclinD1,which is related to HepG2cell proliferation by concentration and duration, and promote the cell proliferation.2.Apelin-13can inhibit the expression of Caspase3,which is related to HepG2cell apoptosis by concentration and duration, and inhibit the cell apoptosis.
