ObjectiveCurcumin(Curcumin,Cur)is a polyphenolic compound extracted from the rhizomes of traditional Chinese medicine such as ginger plants.It is yellow crystalline powder,bitter,insoluble in water,soluble in organic solvents,commonly used food additives and colouring agent,with anti-oxidation,anti-inflammatory,anti-atherosclerosis,anti-microbial,anti-mutation and so on.Since 1995,Menon et al the first time put forward the possibility of anti-tumor of curcumin,a large number of clinical and clinical experimental studies have been confirmed that the anti-cancer effect of curcumin.Studies have shown that curcumin can inhibit gastric cancer,colon cancer,breast cancer,cervical cancer,prostate cancer and other malignant tumors.Because of the high degree of malignancy in hepatocellular carcinoma,and the early detection of difficulties,the low operative value of advanced hepatocellular carcinoma and the poor effect of chemotherapy and the side effects of chemotherapy,a large number of scholars at home and abroad on the mechanism of curcumin treatment of liver cancer done a lot of research,but the specific mechanism is not yet clear.In this study,we used SMMC-7721 cell as the object of this study to research:(1)Effect of curcumin on proliferation and apoptosis of human hepatocellular carcinoma cells.(2)Effect of curcumin on endoplasmic reticulum stress and its corresponding signal pathway.(3)Effects of endoplasmic reticulum stress on proliferation and apoptosis of human hepatocellular carcinoma cells caused by curcumin.Our aim is to look for natural,non-toxic or low-toxic,effective drugs for the treatment of liver cancer.For the curcumin anti-tumor mechanism and the treatment of human liver cancer to provide data and theoretical support.Methods:1.The cells were treated with different concentrations of curcumin(control,2.5?M,5?M,10?M,20?,50?M)for 12 h,24h,48 h.And then MTT assay was used to detect the viability of hepatocarcinoma cells,and cell flow cytometry was used to detect the apoptosis rate of curcumin treated with hepatocarcinoma cells after 12 hours,and compared with the control group.Toinvestigate the effect of curcumin on the proliferation of hepatocarcinoma cells and the effect on the apoptosis of hepatocarcinoma cells.2.The cells were treated with different concentrations of curcumin(control,2.5?M,5?M,10?M,20?M,50?M)for 12 hours,and then the cells were cleaved and extracted.The expression of endoplasmic reticulum stress-related proteins and apoptosis-related proteins were detected by western blot.To explore the effect of curcumin on the stress pathway of endoplasmic reticulum in hepatocellular carcinoma cells.3.The cells were pretreated with salubrinal(sal)prior to curcumin treatment for 12 h.And then MTT assay was used to detect the viability of hepatocarcinoma cells,and cell flow cytometry was used to detect the apoptosis rate of curcumin treated with hepatocarcinoma cells,and the expression of endoplasmic reticulum stress-related proteins was detected by western blot.And statistically analyzed with curcumin alone.To investigate whether curcumin is effective against hepatocellular carcinoma SMMC-7721 cells through endoplasmic reticulum stress response.Rresults1.MTT assay showed that the absorbance of human hepatocellular carcinoma SMMC-7721 cells was significantly decreased and the viable cells were significantly decreased,which indicated that curcumin could inhibit the proliferation of hepatoma cells by increasing the dosage of curcumin and prolonging the time of action.Compared with the control group,the difference was statistically significant(all p <0.05).2.Detection of Apoptosis by Flow Cytometry showed that the apoptotic rate of hepatocellular carcinoma SMMC-7721 cells increased significantly with the increase of curcumin concentration,and compared with the control group,the apoptosis rate of hepatocarcinoma cells was significantly higher than that of the control group(p <0.05).3.Western Blot showed that curcumin could induce the expression of endoplasmic reticulum stress-related proteins and apoptosis-related proteins of SMMC-7721 cells.As the increase of curcumin concentration,the proteins expression increased gradually.And compared with the control group was statistically significant(all p <0.05).4.The cells were pretreated with sal prior to curcumin treatment for 12 h.Western blot showed that the sal could significantly inhibit the expression of endoplasmic reticulum stress-related proteins.And compared with the control group was statistically significant(p <0.05).5.The cells were pretreated with sal prior to curcumin treatment for 12 h.MTT and flow cytometry were shown that the inhibitory rate of hepatocarcinoma cell proliferation in curcumin treated group was significantly higher than that in SAL pretreatment group(p <0.05),and cell apoptosis rate is more obvious(p <0.05).ConclusionsCurcumin can significantly inhibit the proliferation of hepatocellular carcinoma cells and induce apoptosis of hepatocellular carcinoma cells through endoplasmic reticulum stress pathway,which is time-dose-dependent.And endoplasmic reticulum stress suppressor sal pretreatment of hepatocellular carcinoma cells,further confirmed that curcumin through the activation of endoplasmic reticulum stress response induced hepatocellular carcinoma cell apoptosis.The above conclusions on the clinical study of curcumin,and the treatment of liver cancer to explore are of great significance.
