Objective:1. To explore suitable Yunnan Baiyao and PLGA material, choosethe simple diaphragm making method with experiments.2. Through the experimentalstudy of Yunnan Baiyao mixed PLGA material biocompatibility, and its effect onwound healing.Methods: Part I:1.Preparation of composite materials:Dissolve theYunnanbaiyao with PLGA powder material at different mass ratio (1:5,1:10,1:20) inchloroform, then mix into a paste, and tile on the glass by atural spread. After drying,cut into1.5cm×1.5cm size of composite film, dry cold standby.2. Material static in vitro degradation experiment: Yunnan Baiyao mixed materialPLGA diaphragm in the artificial simulated intestinal fluid, in the temperature insidethe degradation temperature37℃, weekly regular replacement of artificial intestinaljuice.In1,2,3,4,6,8weeks,6points in time, will try out the drying to constantweight loss rate, evaluation of its degradation.Part II: The evaluation of biocompatibility of PLGA modifiedmaterials of biological: acute toxicity test, pyrogen test, hemolysis test, micronucleustest extracts of Yunnan Baiyao PLGA hybrid materials used, operation incisionsurrounding tissue HE staining, Masson staining, hydroxyproline assay to evaluate theYunnan Baiyao mixed PLGA material biocompatibility, and compared with the rubber,discuss its influence on wound healing.Results:1. The mixed material mass ratio in the2-4week fastest degradationrate, degradation of about74-78%,6-8week reduced liberation corrosion.2. The mixed material extract acute toxicity test, pyrogen test negative <0.5℃temperature rises, the hemolysis rate≤5%, micronucleus rate≤5‰. 3. Preliminary animal experiment:①HE staining was visible around the large amount of inflammatory cell infiltration,the proportion of Yunnan Baiyao mixed material PLGA group than in control groupwere less positive rubber;②Display inflammatory reaction:The ratio of mixed material group has significantdifference at the same time and rubber group (P <0.05), there are significantdifferences at the same time comparison between the ratio of material group (P <0.05), the ratio of material group were significantly different in different time (P <0.05);③Masson staining of collagen fiber cut blue green, red muscle fibers and cellulose.In7days we can see the inflammatory cell accumulation, fibroblast proliferation,collagen deposition of collagen in small, slender, sparsely.14days to see large incisioncollagen deposition, thick, dense collagen.Conclusion:1. We can through adjusting the allocated proportion between Yunnanbaiyao and PLGA material,enabling it to have the different degeneration speed, thechoice suits anastomat the proportion of request.2. Yunnan Baiyao mixed PLGA is a kind of good biocompatibility,non-toxic, biodegradable materials; and can reduce the wound inflammatory response,promote wound healing. The ratio of mixed material degradation rate of1:5was faster,and the hemolysis rate and micronucleus rate was the lowest, the production of thisratio may be more applicable to a stapler.
