This dissertation is composed of five chapters. The first chapter discussed the antioxidantcapacity of Saururi herba and α-glucosidase inhibitory activity of Saururi herba and Fructus Actinidiachinensis Planch in vitro. The second chapter included the hepatoprotective effect of Saururi herba onacute liver injury induced by carbon tetrachloride in mice. The third chapter studied the hypoglycemiceffect of Fructus A.chinensis on alloxan-induced diabetes in mice. The fourth chapter analysed thefat-soluble components in Fructus S.chinensis by GC-MS. The fifth chapter summarized the biologicalactivity of Fructus A.chinensis.Chapter Ⅰ Bioactivities of Saururi herba and Fructus Actinidia chinensis Planch in virto1. Antioxidant activity of different extracts of Saururi herba was evaluated by scavenging1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and [2,2′-azino-bis(3-ethylbenzothiazoline)-6-sulphonic acid]diamonium salt (ABTS) radical in vitro, in order to provide theoretical basis for activity study in vivo. Thestudy discovered that n-BuOH extract from Saururi herba showed higher DPPH radical scavengingactivity (IC50=16.94μg·mL-1) than that of BHT (IC150=18.71μg·mL-) as positive control, and lower that ofPG and BHA (IC50=0.89and3.2μg·mL-1) as positive controls. ABTS radical scavenging activity(IC50=12.90μg·mL-1) was much lower than that of PG and BHA (IC50=0.81and1.95μg·mL-1), andslightly lower than that of BHT (IC50=7.72μg·mL-1). Petroleum ether extract and EtOAC extract showed lower DPPH and ABTS radical scavenging activity than that of PG, BHT and BHA, which showed thatn-BuOH extract had good antioxidant activity. To study the relationship between concentration andscavenging rate, we discovered that the scavenging activity of DPPH and ABTS radical was concentrationdependent.2. By establishing α-glucosidase inhibitory model in vitro, the α-glucosidase inhibitory activitiesof different extracts from Saururi herba and Fructus A.chinensis were screened in96-well plates, and therelationship between inhibitory rate and extract concentration was also studied. Results showed thatpetroleum ether, EtOAC extract and n-BuOH extract from Saururi herba and Fructus A.chinensis hadgood inhibitory activities. EtOAC extract from Saururi herba had the highest inhibitor activity (IC50=122.7μg·mL-1), and followed by petroleum ether extract and n-BuOH extract (IC50=203.3and659.9μg·mL-1).Petroleum ether extract from Fructus A. chinensis had the highest inhibitor activity (IC50=57.8μg·mL-1),and followed by EtOAC extract and n-BuOH extract (IC50=84.7and124.7μg·mL-1). The activities werehigher than that of Acarbose (IC50=1103.01μg·mL-1) as positive control, and α-glucosidase inhibitoryactivity had positive correlation with mass concentration, their activities were concentration dependent.Chapter Ⅱ Hepatoprotective effect of Saururi herba on liverBy intraperitoneal injection carbon tetrachloride to establish acute liver injury model in mice, thelevels of GOT, GPT, SOD and MDA were detected to evaluate protective effects of petroleum ether extractand n-BuOH extract from Saururi herba. Results showed that petroleum ether extract and n-BuOH extractcould significantly reduce the levels of GOT and GPT in mice serum, significantly increase the level of SOD and reduce the level of MDA in liver homogenate, enhanced antioxidative capacity, which showedthat petroleum ether extract and n-BuOH extract were useful to protect CCl4-induced acute liver injury andhepatoprotective mechanisms was related to improve the antioxidant capacity of liver cells.Chapter Ⅲ Hypoglycemic effect of Fructus Actinidia chinensis PlanchThe diabetic mouse model were established by intravenous injection of alloxan, the levels ofblood sugar, hepatic glycogen and TG, TC, MDA and SOD in serum were detected to evaluatehypoglycemic effects of petroleum ether extract, EtOAC extract and n-BuOH extract from FructusA.chinensis. Results showed that the three extracts had the effects of reducing blood sugar, regulating lipidmetabolism and enhancing antioxidant defense system, which showed that A.chinensis had certain curativeeffect on diabetes. Low dose group of petroleum ether extract and high dose group of EtOAC extract couldsignificantly decrease postprandial blood glucose and fasting blood glucose, medium dose group of EtOACextract and low dose group of n-BuOH extract had better effects on decreasing postprandial blood glucose,medium dose group of n-BuOH extract had better effects on decreasing fasting blood glucose. High dosegroup of petroleum ether extract, three dose groups of EtOAC extract and low dose group of n-BuOHextract could significantly increase the content of liver glycogen. Three extracts could significantlyincrease the content of TC, high and low dose goups of petroleum ether extract and n-BuOH extract, highdose group of EtOAC extract could significantly increase the content of TG. Three dose groups of the threeextracts could significantly increase the level of SOD, three dose groups of petroleum ether extract andn-BuOH extract and high dose group of EtOAC extract could significantly decrease the content of MDA. From the above results could be seen, Fructus A.chinensis presented anti-diabetic effect, the mechanismwas probably concerned with promoting hepatic glycogen synthesis and inhibiting excessive degradation,rectifying the derangement in lipide metabolism and strengthening antioxidant defense system.Chapter IV Analysis of fat-soluble components in Saururus chinensis (Lour.) Baill by GC-MSThe fat-soluble components of petroleum ether extract from S.chinensis were analyzed byGC-MS.15compounds from petroleum ether extract were identified, accounting for43.77%of the totalfat-soluble components. Furans (13.51%), alcohol (6.42%), aldehydes (4.54%), fatty acids (6.4%) and itsmethyl esters (4.75%) were dominant in the fat-soluble components, The main compounds were[2S-(2α,3,4α,5)]-2,5-bis(3,4-dimethoxyphenyl)tetrahydro-3,4-dimethyl-Furan (10.97%),4-dihydro-1-(di-phenylmethylene)-5-hydroxy-4-oxo-Naphthalene (6.93%), n-Hexadecanoic acid (3.51%) and-Sitosterol(3.45%).Chapter V Research progress of biological activity of Fructus Actinidia chinensis PlanchThe biological activity research progress of anticancer effect, antihypeflipidemia effect,resistance mutation and distortion, detoxication, antioxident activity, hepatoprotective effect, enhancingimmune function and boosting intestinal movement were summarized.
