Objective:1.To explore effect of pomegranate peel polyphenols on human prostatecancer and its possible mechanism in vivo and in vitro.2. To study the anticancer activityand apoptosis effective of Pomegranate peel polyphenols or it combined with tamoxifenon human breast cancer MCF-7/Adr and MCF-7cells. Methods: Methyl thiazolyltetrazolium (MTT) assay was used to detect the proliferation of PC-3cells, the cellapoptosis and changes of cell cycle of PC-3cells were analyzed by using flow cytometryafter treated with72h. Use the up-down method to assay the LD50of the pomegranate peelpolyphenols on nude mice, established the PC-3nude mice xenograft models, observe thedifferent dosage of pomegranate peel polyphenols the therapeutic effect on xenografts innude mice, organs and tissue pathology were observed; apoptosis was detected by TUNELassay. VEGF and TNF-α was detected by ELISA. MTT assay was used to detect theproliferation of pomegranate peel polyphenols or it combined with tamoxifen on humanbreast cancer MCF-7/Adr and MCF-7cells, the cell apoptosis of MCF-7/Adr and MCF-7cells were analyzed by using flow cytometry after treated with72h. Results: Pomegranatepeel polyphenols significantly inhibited the cell proliferation of PC-3cells in dose andtime dependent manners, the results of flow cytometry showed that pomegranate peelpolyphenols can arrested in cell cycle progression at G1phase, and obviously induce theapoptosis of PC-3cells. Nude mouse experiments showed that the pomegranate peelpolyphenols can inhibited the volumes of prostate cancer xenografts compared with thecontrol group, and the tumor weights were also decreased, high dosage group(P<0.05),effect of pomegranate peel polyphenols on tumor inhibition ratio was41.66%、36.57%、31.89%. But pomegranate peel polyphenols can cause the toxicity on organs at higherdosage; TUNEL staining revealed that the rate of cell apoptosis increased significantly inpomegranate peel polyphenols groups(P<0.05), and it can improve the TNF-α in serum of nude mice(P<0.05), decrease the middle and high dosage group VEGF in serum ofnude mice. Pomegranate peel polyphenols or it combined with tamoxifen significantlyinhibited the cell proliferation and induce the apoptosis of MCF-7/Adr and MCF-7cells indose and time dependent manners, but the effect of Pomegranate peel polyphenolscombine with tamoxifen highest than use alone. Conclusion: Pomegranate peelpolyphenols could inhibit the proliferation and induce apoptosis of PC-3cells,pomegranate peel polyphenols can inhibit the growth of xenograft by inducing apoptosisand improve the TNF-α, decrease VEGF in serum. Pomegranate peel polyphenols couldinhibit the proliferation and induce apoptosis of MCF-7/Adr and MCF-7cells, and theeffect of Pomegranate peel polyphenols combine with tamoxifen highest than use alone.
