Objective: Medicine mulberry (M.nigra.Linn.) is a unique rare expensive medicinalfruit mulberry germplasm resources in Xinjiang and the Middle East. Medicine mulberryas Uighur medicinal herbs used in the clinical history, but there was no systematic study ofthe chemical composition reported, the lack of quality control standards andmethods.Uighur medicinal ministerial standards included Medicine mulberry herbsmedicine preparations, as the SIPA accordance Maizi He sub oral solution, but have notreceived contained herbs standards belong to varieties upside down.ChinesePharmacopoeia (2010edition) included the traits identification of Medicine mulberry(Morus.alba.Linn),but no TLC and Determination items. In this study, based on theguiding principles of the Chinese Pharmacopoeia, in the course of the study, using avariety of state-of-the-art technology, establish the microscopic identification and the TLCidentification of Medicine mulberry, the determination of moisture and ash, determinationitems, based on it to writing Medicine mulberry quality standards draft. Method:1. Usingmicroscopic characteristics identify Medicine mulberry.2. To establish the thin layerchromatogram identification method for Medicine mulberry flavonoids and anthocyaninscompounds by TLC.3. To establish the method for the determination of the chemicalcomposition of the Medicine mulberry by HPLC.Using a measured multi-ratingextrapolate the content of the sample.4. Multiple batches of Medicine mulberry moistureand ash value were measured.5. UV method and thin layer since the enhancement ofmedicine mulberry extract to determine the activity. Result:1. Powder microscopiccharacteristics of the Medicine mulberry containing endocarp stone cells, calcium oxalatecrystals, clusters of calcium oxalate, non-glandular hairs, light yellow crystallinesubstance, the milk ducts, conduit and seed coat epidermal cells.2. In TLC identification,with silica gel thin layer and microemulsion thin layer methods, respectively. In silica gel thin layer, separation of flavonoids and separation of anthocyanin reference substanceiswere good, but the sample was not very good separation of those,and that pigmentaccumulation near the origin; In microemulsion thin layer, the separation of flavonoidsand anthocyanin were good, and anthocyanins in the sample separation better dispersionand pigments to the top.3. According to the literature, rutin, isoquercitrin, quercetin andchlorogenic acid as indicators to establish a method for the determination of medicinemulberry,and a systemic study of sample preparation and methodology validation; samplemeasurement results: the content of rutin not less than115μg·g-1, of isoquercitrin not lessthan66μg·g-1, of chlorogenic acid not less than155μg·g-1, of quercetin is not lowerthan13μg·g-1.4. Determination of moisture content of12batches of Medicine mulberryan average of12.17%, an average of the total ash was7.71%.5. To establish thefingerprints of medicine mulberry, and research methodology; precision, stability andrepeatability RSD are up to the mustard. Meanwhile established a measuredmulti-assessment method of medicine mulberry; to rutin the internal standard, the relativeisoquercitrin, chlorogenic acid and quercetin correction factors were1.446,1.812,1.837,with the measured values of the external standard method no significant difference.6.Antioxidant activity assay showed that the medicine mulberry to IC50of the DPPH,·OH,O2-· is5mg·mL-1,23mg·mL-1,26mg·mL-1. The thin-layer bioautography to identifythe chemical composition of rutin have antioxidant activity. Conclusion:1. This study inaccordance with the requirements of the guiding principles of the Chinese Pharmacopoeia,the establishment of the microscopic characteristics of the medicine mulberry, TLC,content determination, moisture and ash completed medicine mulberry quality standards.2.In the course of the study, the combination of active and TLC methods, not only can getthe total antioxidant activity against oxidation of the active ingredient can also identify.3.Fingerprinting conducted a preliminary study and to establish the methodology to meet therequirements.4. Smaller difference between the content of the measured results of a test ofmultiple assessment methods projections content of the sample and external standardmethod, can be achieved in the case of a shortage of reference substance to determine thecontent of the multi-index components, and become a new model for traditional Chinesemedicine characteristics of multi-indicators of quality evaluation.
