Objective:To analysis the microRNAs expression profiles in liver of Echinococcusmultilocularis infected mice; explore the expression of miR-106b-25microRNA cluster inthe Echinococcus multilocularis (E multilocularis) infected mice liver. Methods:FemaleBalb/c mice were randomly divided into the experimental group, control group andnormal group. The experimental group was injected with E multilocularisintraperitoneally, the control group was injected with corresponding volume of normalsaline. The normal group was untreated and sacrificed at the same day. Five mice fromthe experimental group and Five from the control group were sacrificed at30,90and180days after injection. The liver samples of3groups were conllected and performed withtotal RNA extraction. The microRNA samples of mice livers infected with Echinococcusmultilocularis (E multilocularis) for3months were hybrided with miRNAs microarraychips. miRNAs expression profiles of hepatic echinococcosis were obtained after dataanalysis, The relative expression levels of miR-93, miR-106b, miR-494, miR-152,miR-191and miR-199a in each group were measured with RT-qPCR at1,3and6months after injection, finally the target genes of miR-93and miR-106b were predictedand analysised by softwares. Results: According to the Gene chip results, compared withcontrol group and normal group,46miRNAs were differentially expressed in the livertissues of experimental group mice,29were upregulated, and17were downregulated;19microRNAs were upregulated more than2times,2microRNAs were downregulatedmore than2times. RT-qPCR showed that the miR-93and miR-106b relative expressionlevels in the samples of experimental group at90days were3.7045±1.767and5.901±2.915respectively and significantly increased by comparing with control and normal groups(P <0.05). The relative expression levels of miR-93and miR-106b inexperimental group at30and180days were unchanged (P>0.05). Bioinformaticsanalysis showed that p21, the common target gene of miR-93and miR-106b, may beinvolved with cell apoptosis and cell cycle regulation. Conclusion: The miRNAsexpression profiles in Echinococcus multilocularis (E multilocularis) infected mice liverwere significantly changed, prompt miRNAs may participate in the process ofEchinococcus multilocularis liver injury, The differential expressions of miR-93andmiR-106b were found in the mice liver infected with Echinococcus multilocularis (Emultilocularis), which may indicate miR-106b-25microRNA cluster is involved inhepatic injury induced by Echinococcus multilocularis.
