Objective: To investigate the expression of Her-2/neu and Smad7in gastric carcinoma discuss itsclinical significance and their correlation, and to investigate the consistency of Her-2/neu expression by fluorescence in situ hybridization (FISH) and immunohistochemistry(IHC).Methods: Selected120cases of gastric adenocarcinoma and50cases of adjacent tissue to analysis theexpression of Her-2/neu and Smad7’s clinical significance and their correlation by IHC, Made the specimeninto tissue microarray, detect the Her-2/neu gene amplification by FISH..Results:77cases Her-2displayed a different staining results in120cases of gastric carcinoma by IHC,showed16cases(3+),6cases of multifocal(3+),37cases(2+),18cases(1+). The positive rate is18.3%. Theexpression of Her-2/neu was not correlated with the patient’s gender, age, ethnicity(P﹥0.05),but wasassociated with the site of the tumor, degree of differentiation, Laurens parting correlation(P﹤0.05),41cases were positive by FISH. All five FISH-positive cases with the IHC multifocal (3+) were comparedspecimens of different coloring strength parts by FISH, all five cases were amplification.. IHC and FISH inline with the rate of91.5%. Kappa coefficient was0.960(P﹤0.001).101cases Smad7were positiveexpression,the positive rate was84.16%. The expression of Her-2/neu was correlated significantly withSmad7in gastric carcinoma(P﹤0.05).Conclusion:1. Using tissue microarray detected by IHC and FISH coincidence rate (91.5%), highconsistency (Kappa coefficient=0.960, P<0.001).2Patients with intestinal type GC and gastro esophagealjunction tumors may be the potential responders to trastuzumab combined with chemotherapy therapy.3Joint evaluation of Her-2and Smad7to provide significant value to determine the prognosis of patientswith gastric cancer.
