Objective:Study on the effect and mechanism that small heat shock proteins Hsp16.3of Mycobacteriumtuberculosis reacts at the apoptosis of macrophages.Methods:1Infectious status of macrophages was observed with confocal laser scanning microscopy(CLSM),which were infected by international standard virulent strain of mycobacterium tuberculosisH37Rv strains (H37Rv)、 lack of heat shock proteins Hsp16.3gene deletion strains ofinternational standard virulent strain of mycobacterium tuberculosis H37Rv strains (△H37Rv)、BacilleCallmette-Guerin(BCG) and lack of heat shock proteins Hsp16.3gene deletion strains of BCG (△B CG).2After macrophages were infected1h、3h、6h、12h,flow cytometry was used to detect macrophagesapoptosis rate, Caspase-3and Bcl-2expression were examined by Western Blot.Results:1The difference of macrophages apoptosis rate is related to mycobacterium tuberculosis’svirulence, macrophages apoptosis rate of BCG group were higher than H37Rv group, macrophagesapoptosis rate of△BCG groupwere higher than△H37Rv group.Macrophages apoptosis rate of smallheat shock proteins Hsp16.3gene deletion strains`s group were higher than normal strains, macrophagesapoptosis rate of△H37Rv group and△BCG groupwere higher than H37Rv group and BCG group.2Macrophages infected by H37Rv、BCG、△H37Rvand△B CG can induce expressCaspase-3andBcl-2.With the infected time increasing, the expression of Caspase-3of H37Rv group、BCG group、△H37Rv group and△B CG groupwas also increasing,the expression of Caspase-3was highest at12h, theexpression of Caspase-3of BCG group was higher than H37Rv group, the expression of Caspase-3of△B CG groupwas higher than△H37Rv group; The expression of Caspase-3was no difference betweenH37Rv group and△H37Rv group, BCG group and△B CG group.With the infected time increasing, theexpression of Bcl-2of BCG group and△BCG groupwas also increasing,the expression of Bcl-2washighest at12h,however,the expression of Bcl-2of H37Rv group and△H37Rv groupwas decreasing, theexpression of Bcl-2was lowest at12h; the expression of Bcl-2of BCG group was higher than△BCGgroup, the differences had statistical significance at3h、6h、12h(P<0.05), the expression of Bcl-2of H37Rvgroup was higher than△H37Rv group, the differences had statistical significance at1h、3h、6h(P<0.05).Conclusions:1Infected macrophages apoptosis rate was related to mycobacterium tuberculosis’ virul-ence, macrophages apoptosis rate of the less virulent mycobacterium tuberculosis was more notable. Smallheat shock protein Hsp16.3can affect Infected macrophages apoptosis rate, macrophages apoptosis ofsmall heat shock proteins Hsp16.3gene deletion strains`s group were higher than normal strains.2Macrophages infected by mycobacterium tuberculosis can induce express Caspase-3and Bcl-2,the expression of Caspase-3and Bcl-2was related to mycobacterium tuberculosis’ virule-nce. Compare small heat shock proteins Hsp16.3gene deletion strains`s group with normal strains,theexpression of Caspase-3and Bcl-2was different that may be related to whether mycobacteriumtuberculosis express small heat shock protein Hsp16.3or not.3Small heat shock protein Hsp16.3of Mycobacterium tuberculosis could regulate apoptosis ofinfected macrophages,which may be realized by facilitating to express Bcl-2,and it’s with nothing withCaspase-3.
