Objective To discuss the expression of CCAAT/enhancer binding protein alpha (C/EBPa) protein in cervical carcinoma tissue and its effect to tissues cellular proliferation in cervical squamous cell carcinoma.Methods The expressions of C/EBP alpha gene mRNA and Ki-67mRNA level were detected by RT-PCR in12cervical tissues of Uygur women in Xinjiang (5normal cervical tissue and7cervical carcinomas). Using immunohistochemistry method to detect the expression of C/EBP alpha protein, Ki-67protein and PCNA protein in84cervical tissues of Uygur women in Xinjiang (37chronic tissue and47cervical cancer tissue). Statistic the correlation between C/EBP alpha gene and Ki-67gene and between C/EBP alpha gene and PCNA gene through the application of statistical methods in cervical carcinoma tissues and chronic cervicitis tissues.Result (1) C/EBP alpha gene expression at mRNA level in normal cervical tissues is higher than those of cervical squamous cell carcinoma tissues.(2) Ki-67gene expression at mRNA level in corresponding C/EBP alpha cervical squamous cell carcinoma tissues is higher than those of corresponding normal cervical tissues.(3) C/EBPa protein expression level in chronic cervicitis tissues is higher than cervical squamous cell carcinoma tissues, the difference has statistic significance, P<0.001.(4) Ki-67protein expression in corresponding C/EBP alpha chronic cervicitis tissues is lower than in corresponding C/EBP alpha cervical squamous cell carcinoma tissues, and the difference has statistic significance, P<0.05.(5) PCNA protein expression in corresponding C/EBP alpha chronic cervicitis tissues is lower than in corresponding C/EBP alpha cervical squamous cell carcinoma tissues, and the difference has statistic significance, P<0.001.(6) The relationship between C/EBP alpha protein expression and Ki-67protein expression was negatively correlated in cervical cancer tissues (P<0.05); The relationship between C/EBP alpha protein expression and PCNA protein expression was negatively correlated in cervical cancer tissues (P<0.05)Conclusion The expression of C/EBP alpha gene mRNA and protein in cervical cancer tissue is reduced. The C/EBP alpha gene could cause the inhibition of tissues cellular proliferation in cervical squamous cell carcinoma.
