| ObjectiveTo explore the effect of pure realgar on apoptosis of gastric adenocarcinoma cell line SGC-7901.MethodsGrowth inhibition ratio of SGC-7901cells treated by different concentrations of pure realgar was assayed by3-(4,5)-dimethylthiahiazo(-z-yl)-3,5-di-phenytetrazoliumromide (MTT).The apoptosis of SGC-7901cell were detected by Annexin V/PI double staining method and flow cytometry. The expression of Bcl-2and Bax protein were assayed by Western blot.ResultsPure realgar inhibited the SGC-7901cells proliferation and introduced apoptosis in a dose dependent manner. It could also dose dependently induce early apoptosis. Furthermore, pure realgar reduced the ratio of Bcl-2/Bax by down-regulating the expression of Bcl-2protein and up-regulating the expression of Bax protein.ConclusionPure realgar has the potential of inhibiting the proliferation and inducing the apoptosis in SGC-7901cells. The mechanism may be related to reducing the ratio of Bcl-2/Bax. |
