objective: To observe the expression of integrin β3and Akt phosphorylation(p-Akt) by making rat model of abdominal aortic aneurysm and explore themolecular mechanisms of abdominal aortic aneurysm.Methods: Making a rat model of abdominal aortic aneurysm and measuringthe diameter of all aortas then calculated their extended rate of the aorta. HE stainingwas used to observe the change of pathology. Immunohistochemistry, westernblotting and real time PCR were used to detect the expression of p-Akt、t-Akt、p-ERK、t-ERK and integrin β3from the level of protein and gene.Results:Rat model of abdominal aortic aneurysm was successfully made.Compare to saline group and normal group, the extended rate was obvious rising andthe infiltration of inflammatory cell was more apparent. Immunohistochemistryshowed that the expression of integrin β3、p-Akt and p-ERK in experimental group、saline group and normal group were (0.33±0.07) vs (0.20±0.06) vs (0.19±0.07)(P<0.05),(0.247±0.046) vs (0.160±0.041) vs (0.167±0.065)(P<0.05) and(0.136±0.047) vs (0.141±0.027) vs (0.148±0.235)(P>0.05); real time PCR showedthe expression of integrin β3mRNA in experimental group、saline group and normalgroup were (2.29×105±2.67×105) vs (2.69×104±2.43×104) vs (1.19×104±1.36×104); western blot showed the expression of p-Akt and p-ERK in experimentalgroup、saline group and normal group were (0.559±0.134) vs (0.115±0.063) vs (0.141±0.107)(P<0.05) and (0.498±0.085)vs (0.581±0.095) vs (0.539±0.109)(P>0.05).Conclusions: The increasing expression of integrin β3and p-Akt mayparticipate in the occurrence and development of experimental abdominalaneurysms.
