Objective:To observe the effect of shenhuahuakeli on expression of a-SMA and MMP-9in atherosclerosis plaque of ApoE knockout mice for exploring its plaque stabilizing action and possible mechanism. Methods:sixty ApoE knockout mice,6-8weeks old,were fed with high-fat diet for12weeks. Another fifteen C57/BL mice were fed with commom diet for normal group, HE stain to comfirm that mature atherosclerotic plaque has being formed, then animals were randomly into five groups,12in Each group. They were continuously fed with high—fat diet and to the different dose of shenhuanghua—treated groups, respective drugs were given for another12weeks. By the end of the experiment, all mice were sacrificed. The morphology and composition of atherosclerotic plaques in aortic roots were examined with HE stain. Additionally, the expressions of a-SMA, MMP-9in plaque was determined by the immunohistochemistry method. Results:After treatment for12weeks, shenhuanghua significantly inhibited the expression of MMP-9(P<0.05), whereas the expression of a-SMA in plaque was higher (P<0.01) than that in the model group. Conclusion:shenhuanghua can dose-despensible significantly decrease the frequency of plaque rupture in aorta of ApoE knockout mice and do favour to plaque stability, and the mechanism is possible that shenhuanghua decrease level of plasma lipid and thicken the fibrehat of vulnerable plaque,in other way, shenhuanghuakeli can also decrease inframmation effect.
