Objective: To investigate the reversal effects of the CIK on A549/DDP cell line. Toinvestigate the impact of CIK on cell cycle and apoptosis.Methods: A549and A549/DDP cells were cultured with CIK,DDP alone or incombination with CIK respectively. The inhibition of DDP and the inhibition of DDP incombination with CIK were assessed with MTT and the IC50values werecalculated..Respectively,A549/DDP alone or combined with CIK were cultured for48h.Anenxin V-PI staining was applied for analysis of A549/DDP cell line apoptosis andcell cycle.Results: The IC50was5.9μg/ml、31.7μg/ml when A549and A549/DDP cells treatedwith cisplatin. The difference between the two groups had statistical significance (P<0.05).Chose CIK:A549/DDP=15:1value as its non-toxic dose. cisplatin combined with CIK thatbellowed its IC10value could significant decrease the IC50value to11.23μg/ml onA549/DDP cells. RI=5.2, RF=2.7。 CIK cells markedly affected the cell cycle ofA549/DDP cell line, which was bolcked at G1stage when they were cultured together.CIKcells significantly induced A549/DDP cell line apopotosis compared with control.Conclusion: CIK can enhance the sensitivity of A549/DDP cells to cisplatin. CIKcells markedly affected the cell cycle of A549/DDP cell line, which was bolcked at G1stage and CIK can induce cell apoptosis in a populatin dependent manner and can enhancethe sensitivity of A549/DDP cells to cisplatin.
