Objective:1.To establish and evaluate models of mice with allergic rhinitis.2.Toinvestigate the expression and significance of aquaporin5in mice nasal mucosa withallergic rhinitis.Method:1.The animal models of mice with allergic rhinitis was established by thegeneral and local sensitization with ovalbumin.The animal model with allergic rhinitiswas evaluated by behavioral score. The levels of specific IgE and IL-4in peripheralblood was detected by enzyme-linked immunosorbent assay. The expression ofeosinophils was observed in nasal mucosa under the microscope by HE staining.2.The expression of mRNA of aquaporin-5in nasal mucosa was detected by RT-PCR.Result:1.Behavior scores of all mice in the experimental group were more than5points, significantly higher than the control group. The level of specific IgE and IL-4of experimental group in peripheral blood was higher than controlgroup.Hematoxylin-eosin staining showed that the cilia in nasal mucosa wasoff,interstitial edema,vasodilatation and a large number of eosinophil, lymphocytesand plasma cells were soaked.2.The mRNA of aquaporin-5in mice nasal mucosawith allergic rhinitis was significantly higher in normal mice.Conclusion:1.The model of mice with allergic rhinitis can be successfullyestablished by the general and local sensitization with ovalbumin.2.The expression ofaquaporin-5in mice nasal mucosa with allergic rhinitis are higher than normal mice,suggesting that aquaporin-5may be involved in the pathogenesis of allergicrhinitis,which cause nasal hypersecretion of liquid.
