Research Background:The incidence of head and neck squamous cell carcinoma(HNSCC)is the sixth highest in the world.Laryngeal squamous cell carcinoma originating from laryngeal epithelial cells has a second highest incidence in head and neck squamous cell carcinoma,and is particularly common in northern China.Laryngeal squamous cell carcinoma(LSCC)is a highly invasive and metastatic tumor whose malignant biological behavior is the main cause of clinical treatment failure.The survival prognosis of patients with LSCC has not been significantly improved.Therefore,exploring the molecular mechanisms of LSCC's malignant biological behavior and finding new therapeutic targets may contribute to the early prevention,diagnosis and prognosis of LSCC.Deregulating Cellular Energetics is one of the top ten characteristics of tumor cells.Fatty acids play an important role in energy metabolism.Abnormal expression of fatty acid metabolism-related genes in tumor cells may lead to abnormal energy metabolism of tumor cells and promote the development of tumor cells.In the previous study,our group found that the fatty acid metabolism related gene acetyl-CoA acyltransferase 1(ACAA1)gene was abnormal in transcript level and protein level of LSCC by real-time PCR and immunohistochemical staining,which suggests that it may affect the proliferation and metastasis of LSCC by regulating fatty acid metabolism.Research purposes:To explore the effect of abnormal expression of fatty acid metabolism related gene ACAA1 in LSCC on its malignant biological behavior,and provide theoretical basis for further experimental research.Research method:1.Using the tumor genome database created by the National Center of Biotechnology Information(NCBI)to extract the messenger RNA(mRNA)level sequencing data of head and neck squamous cell carcinoma for bioinformatics analysis and statistics.To investigate the expression of the ACAA1 gene in human HNSCC.2.To establish an exogenous overexpressing ACAA1 LSCC cell strain,and to verify the effect of ACAA1 gene on the biological behavior of LSCC malignant tumor by CCK8 proliferation assay,plate colony formation assay,cell scratch assay and cell invasion assay.Research result:1.Fatty acid metabolism related gene ACAA1 down-regulated expression in LSCC samples.Bioinformatics analysis showed that the transcription level of ACAA1 gene was significantly down-regulated in LSCC samples.The expression levels of ACAA1 in LSCC tissues were down-regulated in combination with the previous study of our group,and the results of bioinformatics analysis were further confirmed the accuracy.2.High expression of ACAA1 gene can inhibit the malignant biological behavior of LSCC other than migration ability.Overexpression of the ACAA1 gene significantly inhibited the proliferation,cloning and invasive ability of LSCC cells,but had no effect on their migration ability.Conclusion:1.The expression level of ACAA1 gene is significantly down-regulated in LSCC;2.Exogenous overexpression of the ACAA1 gene can inhibit malignant biological behavior of LSCC in addition to migration.In summary,the fatty acid metabolism related gene ACAA1 may be a potential candidate tumor suppressor gene for LSCC.
