Objective: By measuring the photosensitizer photofrin-Ⅱconcentration ofesophageal cancer cells and esophageal epithelial cells in vivo and vitro,the studywas to investigate the affinity of tumor cells to the photofrin-Ⅱ,in order to providesthe basis for targeted principle in photodynamic therapy.Methods:1.Cells line detection:high performance liquid chromatography was applied tomesure the photofrin-Ⅱ concentration in immortalized human esophageal epithelialcell line SHEE and its malignant transformation cell line SHEEC, investigate the lawof their absorption.2.Primary cell detection:collecting the esophageal cancer tissue and normal tissueadjacent to carcinoma from esophageal cancer patients who receiving surgerytreatment,adopt the method of primary culture, developped esophageal cancer cellsand esophageal epithelial cells, and then use photofrin-Ⅱ culture them respectively,apply high performance liquid chromatography method to detect the photofrin-Ⅱconcentration of them,compare their differences.3.Cells in vivo detection:collecting the esophageal cancer tissue and normaltissue from esophageal cancer patients who receiving photodynamic therapy,usingimmunomagnetic beads technology to separating the esophageal cancer cells andesophageal epithelial cells, apply high performance liquid chromatography method todetect the photofrin-Ⅱc oncentration of them,compare their differences.Results:1.SHEE and SHEEC was incubated in photofrin-Ⅱafter150minutes, with theextension of incubation time, the absorption of photofrin-Ⅱ in SHEEC is more thanSHEE,there was significant difference(P<0.05).2.At the concentration of30μg/mL of photofrin-Ⅱ,with the extension of incubation time, the absorption of photofrin-Ⅱi nprimary esophageal cancer cells ismore than the primary esophageal epithelial cells,there was significant difference(P<0.05).3.The absorption of photofrin-Ⅱ i nesophageal cancer cells is more thanesophageal epithelial cells who was injected photofrin-Ⅱ.Conclusion: Under the same conditions,the absorption of photofrin-Ⅱ inesophageal cancer cells is more than esophageal epithelial cells, subjected thatphotofrin-Ⅱ has a unique affinity for esophageal cancer cells.
