In this paper, we analyzed the content difference of the principal component isomers inglycyrrhizin preparations, and studied and campared different concent detection methods ofbilirubin in calculus bovis.Chapter One:(1) This chapter describes the research progress of isomers of glycyrrhizinacid, introduces the separation and detection methods of glycyrrhizin acid and its isomers, andclarifies the objective and significance of the study.(2) This chapter describes the researchprogress of bovis calculus, summarizes the detection methods of bilirubin in bovis calculusboth at home and abroad, clarifies the objective and significance of the study.Chapter Two: A new analytical method for the simultaneous determination of isomers ofglycyrrhizin acid and related substances by HPLC was established. Determination methods ofglycyrrhizin acid were compared between the new method and National Drug Standards ofChina. The effects of different mobile phase, column temperature and flow rate on theresolution and sensitivity of each component were discussed. The results showed that theestablished method can separate the principal component isomers efficiently, and thedetection results of the content of the relevant materials were true and accurate. This methodhave good precision and reproducibility, high sensitivity, accurate and reliable results, whichcan be used for the determination and quality control of glycyrrhetate ammonium bulk drugsand formulations of different dosage forms.Chapter Three: The establied HPLC method was used to simultaneously separate anddetermine the principal component of glycyrrhizin in four generations, which were18α-glycyrrhizic acid,18β-glycyrrhizinic acid, related substances A and related substances B.The content difference of various components in the same generation preparations withdifferent dosage form and different generation preparations with same dosage form, theirvariation trends and composition ratio were analyzed. The differences between imported anddomestic various glycyrrhizin preparations were analyzed comparatively. The influences ofdifferent methanol concentration and ultrasonic time on the dissolubility quantity of principal components in glycyrrhizin preparation were discussed. The optimum conditions for samplepreparation were selected. The results showed that when the glycyrrhizin preparation wasdifferent, the content of various components and the composition ratio of principal componentwere different. The method and result which were established by this paper, can be used forselecting clinical medicines, controling quality of medicines, researching new glycyrrhizinpreparations. It may be practical value for the selection of clinical medicine andnew-medicine research. It can provide a basis for supplements to Chinese Pharmacopoeia andamendments to National Drug Standards of China.Chapter Four: A new method for determination the content of bilirubin in calculus bovisby HPLC was established. When the samples were determined by HPLC, the optimum samplepreparation conditions and chromatographic conditions were discussed. The optimum samplepreparation conditions were discussed, when the samples were determined by UltravioletVisible(UV) method which was recorded in the2010Edition China Pharmacopoeia. BothHPLC and UV were used to determining bilirubin in calculus bovis. The determination resultswere compared and analyzed. The content differences of bilirubin in natural and cultivatingcalculus bovis were analyzed and compared, coming from different producing areas, differentkinds and different grinding methods. Results showed that the HPLC method was true andaccurate. The contents of bilirubin in calculus bovis coming from of different producing areas,different kinds and different grinding methods were different. It can provide references forestablishing quality standard, for selecting clinical medicine, for selecting calculus boviskinds in pharmaceutical industry, for selecting raw material and the preparation process inproduction process, for exploiting resources and for reserching new calculus bovispreparations.
