Objective: To prepare hyaluronic acid targeted curcumin chitosannanoparticles and consider the feature of HA-CUR-CS-NPs. To study thevitro cytotoxicity of HA-CUR-CS-NPs on A549cells and HepG2cells.Methods:1Through ionotropic gelation method which combine the negativelycharged ion on TPP with free amino on chitosan to form nanoparticles withcurcumin as antitumor drug and hyaluronic acid were connected to thechitosan by covalent coupling, then hyaluronic acid targeting curcuminchitosan nanoparticles were prepared.2Hyaluronic acid curcumin chitosan nanoparticles was used on A549cells of CD44high expression and HepG2cells of CD44low expression.MTT experiment as used to research the cytotoxicity on two cells and thetargeted of hyaluronic acid curcumin chitosan nanoparticles was evaluatedpreliminary.Results: The optimum formula was as follows: the concentration of CSwas2.5mg·mL-1, the concentration of TPP was2.0mg·mL-1,the mass ratio of CTS/TPP mass ratio was4.5:1,CUR was2mg. The average entrapmentefficiency was42.73±0.30%, the average loading capacities was28.44±0.12%,the average binding rate was38.97±0.53%. The averageparticle sizes of CUR-CS-NPs and HA-CUR-CS-NPs were143.9nm and205.3nm.Under transmission electron microscopy, HA-CUR-CS-NPsdisplay regular shape, uniform distribution of particle size,for the Fouriertransform infxared spectroscopy of HA-CUR-CS-NPs were observed. Theresults of MTT test showed that the proliferation inhibition effect ofHA-CUR-L on A549cells of HA receptors are highly expressed,wasCUR<CUR-L<HA-CUR-L. On HepG2cells it wasCUR<CUR-L≈HA-CUR-L.MTT results, the inhibition rate of CUR-CS-NPs andHA-CUR-CS-NPs on A549cells of high expression of HA receptors arehighly expressed are higher than CUR, and the latter than the former; theinhibition rate of CUR-CS-NPs and HA-CUR-CS-NPs on HepG2cells oflow expression of HA receptors are higher than CUR, and both inhibitionrate is basically the same.Conclusions: To prepare the HA-CUR-CS-NPs by ionotropic gelationtowas simple, exact and convenient. The results of vitro release test showedthat the sustained-release effect of HA-CUR-CS-NPs is the best. The resultsof MTT assay displayed that the inhibition rate of HA-CUR-CS-NPs isbest on A549cells. HA-CUR-CS-NPs significantly enhance the proliferation inhibition effect on tumor cells,and it areanticipated to be promising alternate carriers for targeting drugs....
