| Objective:To establish a CE-ECL (capillary electrophoresis coupled withelectrochemiluminescence) detection for the determination ofcisatracurium besylate and the degradations of cisatracurium besylate inplasma; To develop the indirect CE-ECL method for the determination ofcatecholamines and its metabolites.Methods:1. A CE-ECL method for determination of cisatracurium besylate andthe degradations of cisatracurium besylate in plasma:24mmol/L pH2.9phosphate buffer solution (PBS) containing10%acetonitrile as therunning buffer, a separation voltage of13kV and100mmol/L pH8.5phosphate buffer solution containing2.0mmol/L Ru(bpy)23in detectioncell. Rocuronium was as internal standard (IS). The plasma sample wasinjected into CE after separating protein with perchloric acid. Uniformdesign was applied in CE separation.2. CE with ECL quenching method for the determination ofepinephrine (E) and norepinephrine (NE):30mmol/L pH7.9PBS containing6μmol/L TPA as the running buffer, a separation voltage of17kV; injection voltage of16kV and injection time of5s;100mmol/L pH8.5PBS containing0.5mmol/L Ru(bpy)23in the detection cell.3. An indirect CE-ECL method for the determination of metanephrines(MNs):30mmol/L pH8.5borate buffer solution as the running buffer, aseparation voltage of17kV; injection voltage of16kV and injection timeof10s;100mmol/L pH8.5borate buffer solution containing2mmol/LRu(bpy)23in the detection cell. The MNs was injected into CE afterprecolumn-derivatization with formaldehyde for1h (pH8.5).Results:1. The linearity of concentration for cisatracurium besylate andlaudanosine was0.010~8.0mg/L and0.010~40.0mg/L, respectively.All of the coefficient variation (CV) for migration time and ECL intensitywere less than3%.2. The linearity of epinephrine and norepinephrine was33.0~3333.0μg/L and18.0~3380.0μg/L. This method can be used for thedeterminantion of E and NE in urine.3. The linearity of metanephrines was10.2~1020.0μg/L, and thismethod can be used for the determination of MNs in urine ofpheochromocytoma patients.Conclusions:In this research, we developed the CE-ECL method for determination of rocuronium, cisatracurium besylate, an indirect CE-ECL method fordetermination of CA and MNs, which can be used in urine ananlysis forpheochromocytoma. The CE-ECL method can be applied in clinicallaboratory for pharmacokinetics and biochemical metabolism. |
