Objective: Construct β-catenin micRNA expression vector to study therelationship of hypoxia-inducible factor-1α and the Wnt/β-cateinn signalpathway. Explore HIF-1α can regulate the proliferation and invasion ofgastric cancer cell line SGC-7901through the signaling pathway.Method:SGC-7901cell line was transfected with β-catenin micRNAplasmid, and establish stable transfection with targeted interference ofβ-catenin. RT-PCR analysed the interference effect of stably transfected celllines. The biological characteristics of the control group,liposome group,negative control group, hypoxia group, hypoxia interference group weretested by Doubling time,Plate colony and Invasion assay. Then, there weresix groups: control group, hypoxia group, double hypoxia group, controlRNA interference group, hypoxia RNA interference group, double hypoxiaRNA interference group. RT-PCR and Western blot were used to evaluate changes in HIF-1α, β-catenin, CyclinD1, MMP-7mRNA and protein levelsin the six groups.Results: The gastric cancer SGC-7901cell line of stability interferedthe β-catenin which was constructed.Negative control group and liposomegroup compared with the control group were not statistically significant.Hypoxia group compared with the control group, the growth andproliferation slowed down, the ability of invasion increased; Hypoxiainterference group compared with Hypoxia group, the number of colonyformation、 double time and invasion reducd. Hypoxia group anddouble hypoxia group, HIF-1α, the β-catenin, CyclinD1, MMP-7proteinand mRNA expression was elevated;used RNAi technology targeting withβ-cetenin, HIF-1α, β-catenin, CyclinD1, MMP-7protein and mRNAexpression of hypoxia interference group and double hypoxia interferencegroup were significantly reduced. Compare the double hypoxia interferencegroup minus hypoxia interference group with the hypoxia interferenceminus interference, the mRNA and protein expression level of cyclinD1andMMP-7increased。Conclusion:1)The gastric cancer SGC-7901cell line of stabilityinterfered the β-catenin which was constructed successfully.2)With thehypoxia environment,Beta-catenin at the same time exist in the upstreamand downstream of HIF-1alpha and be able to control each other.3) HIF-1αcan stimulate the activation of Wnt/β-catenin signaling pathway, affect downstream gene CyclinD1and MMP–7and promote the proliferation andinvasion of gastric carcinoma.
