FNBP1is Involved In Morphology Control And Growth Regulation Of Human Hepatocyte7702Cells

Objective: To validate the expression and subcellular localization in7702cells,and trying to clarify the role of FNBP1in morphology controland growth regulation of7702cells by observing the biological effect aftersilence of endogenetic FNBP1.Methods: The expression of FNBP1in7702cells was observed byRT-PCR and Western blot respectively at mRNA and protein levels.thesubcellular localization of FNBP1in7702cells was observed bycolocalization analysis. The biological effects after silence of endogeneticFNBP1were observed by HE stain, XTT and FCM.Result: FNBP1is expressed steadily in7702cells. The subcellularlocalization of FNBP1in7702cells is in the cortex.After silence ofendogenetic FNBP1,the cellular morphology of7702cells changes in tobranched fibrous shape and the proliferation is inhibited.And this change isrecovered when FNBP1is reexpressed.Conclusion: FNBP1plays a important role in morphology control andis involved in growth regulation of human hepatocyte7702cells.
