Objective: This study mainly discusses the protective effect of SijunziDecoction on the scalded Wistar rat’s intestinal barrier and its mechanism ofaction from the perspective of apoptosis. Methods:1. Experimental groups:choose55Wistar rats (provided by the experimental animal center of LuzhouMedical College) one week before starting the experiment with a weight of(250±25) g, male or female; conditions: healthy, put in different cages, roomtemperature at26℃,55%humidity environment, free access to food, water andrandomly divide into three groups:5/55-normal control group,25/55-scaldcontrol group (the scalded group),25/55-JWSJZT treatment group (treatmentgroup). Set12hours,1day,3days,5days, and7days after scald for fivephases; each phase has5rats.2. Chinese medicine preparation: add rhubarb toSijunzi decoction with a proportion as codonopsis: poria: atractylodes: licorice:rhubarb=25g:20g:20g:5g:10g. The crude drug weighs80g and put300mlwater in it. After boiling with gentle fire for30minutes, filter and concentrate itto1.0g/ml, stored at4℃and warmed before taking.3. Modeling: According tothe literature[1],make30%TBSA III degree burns on the back of the scaldedgroup and the treatment group and all Wistar rats experience the same scaldcondition; give immediate anti shock and wound coated silver sulfadiazinecream and put into mouth the gavage needle for the treated group six hours later in wakening state and slowly inject the prepared Sijunzi decoction,2ml onceand twice daily; feed the burned control group and normal control group withthe same amount of normal saline in the same way.4. Specimen collection andhandling: in different phrases, extract separately specimen of approximately3cm and mesenteric lymph nodes (MLNS)2cm away from the ileocecalsegment of the small intestine; fixed by4%paraformaldehyde, dehydrated.Paraffin sections experience HE staining, and Terminal-deoxynucleoitidylTransferase Mediated Nick End Labeling (TUNEL) detection;5. Indexdetection and methods:①Through the light microscope, observe differentgroups of small intestinal epithelial cells histopathological changes in the caseof HE staining, TUNEL staining and make comparisons;②Through the lightmicroscope, observe the mesenteric lymph node lymphocytes histopathologicalchanges in the case of HE staining, TUNEL staining and make comparisons;③Figure out the apoptosis of small intestinal epithelial cells in different phrasesand make statistical analysis;④Figure out the apoptosis of mesenteric lymphnode lymphocytes in different phrases and make statistical analysis. Results:1.the structures in the small intestinal mucosa and mesenteric lymph nodes ofthe control group are not injured, small intestinal mucosa and mesenteric lymphnode tissue of burned Wistar rats at an early age are evidently injured and forthe treatment group, small intestinal mucosa and mesenteric lymph node tissueof Wistar rats are significantly bettered compared with the burned group.2. theintestinal epithelial cells and mesenteric lymphocyte apoptosis of burned Wistar rats increased at each time point, compared with the control group, reached itspeak at ld and then gradually decreased, but all were significantly higher thanthe treatment group (P <0.01). For the treatment group, the epithelial cells ofintestinal mucosa and mesenteric lymphocyte apoptosis rates at each time pointwere separately lower than the burned group at each time point, which is ofstatistical significance(P<0.01). Conclusion:Early in use of Jiawei Sijunzidecoction for rats is able to relieve burns of small intestinal mucosa andmesenteric lymph node tissue damage and resist against the apoptosis of smallintestinal epithelial cells and mesenteric lymph node cells. Apoptosis inintestinal epithelial cells and mesenteric lymphocytes is likely to be animportant reason that the intestinal barrier of burned Wistar rats gets injured.
