| Objective:Gastric cancer is one of the most commonmalignant tumors in China whose morbidity and mortality rate rank thefirst in the various digestive malignancies and it also has bad effect onhuman health. It can enhance the rate of survival for five years if gastriccancer is found early and operated immediately. However, most of thepatients of gastric cancer are diagnosed in their later period andtherapeutic effect is far from satisfactory. So far, a large number ofresearchers focus on the study of pathogenesis and the process of gastriccancer and one of the heated study is the research on the abnormalmechanism of apoptosis on cell in gastric cancer.The occurrence anddevelopment of gastric cancer is still the focus of the present study..Apoptosis is an important mechanism to adjust the balance of the bodytissues, the process is subject to strict control. Cells by the loss ofadhesion and hypoxia apoptotic stimuli such as DNA damage, cell cyclearrest, mtochondrial apoptosis-related active substances are released frommediated apoptosis, these active substances cytochrome C,apoptosis-inducing factor (apoptosis inducing factor AIF) endonuclease G(EndoG) the Bitl (Bcl-2inhibitor of transcription), etc., which, byAIF-mediated apoptosis and Bitl caspase (half cystine protease) non-dependent apoptosis. About gastric tissue AIF Bit1expression oftheir mutual relationship little studied. In this study,immunohistochemistry (SP method) to detect AIF Bit1expression ingastric carcinoma and normal gastric mucosa and in healthy volunteers,and analyze its relationship with the biological behavior of gastriccarcinoma, and to explore in gastric carcinogenesis, infiltration andmetastasis.Methods To select49cases of biopsy specimens, including19cases ofpatients with early gastric cancer,21cases of atypical hyperplasia, andnine cases of normal gastric mucosa specimens. All cases were fromDepartment of Pathology, Affiliated Hospital of Luzhou Medical Collegefrom Jan2008to Dec2012hospital of gastric cancer in high-risk groupscensus endoscopic biopsy specimens, including24males and25femalepatients. Selected34cases of biopsy specimens, including18males and16females, were shaped cell carcinoma of the histological type; agerange48to76years, with an average age of61.1±7.14years. Allpatients who underwent preoperative chemotherapy and radiotherapywere confirmed by endoscopy biopsy or surgical resection ofhistologically proven gastric cancer. Immunohistochemistry (SP method)in gastric carcinomas and normal gastric mucosa and mucosa adjacentgastric AIF and Bitl protein expression, analysis of both expression andgastric cancer patients’ gender, age, degree of differentiation, clinical stage clinical biology behavior relationship, and to further explore therelationship of AIF and Bitl of expression with lymph node metastasis ingastric. All application SPSS13.0statistical software for statisticalanalysis (P <0.05), there are significant differences.Results AIF over expression rate of early cancer, dysplasia and normalgastric mucosa are84.2%(16/19),71.4%(15/21) and44.4%(4/9). Theanalysis shows that the AIF in early cancer over expression is higher thanin normal gastric mucosa (P <0.05). Compared with atypical hyperplasia,there is no significant difference (P>0.05); AIF over expression in thedysplasia with normal gastric mucosa shows no significant difference (P>0.05). Bitl in early cancer, dysplasia and normal gastric mucosa, thepositive rate are78.9%(15/19),85.7%(18/21) and44.4%(4/9). Theanalysis shows Bitl atypical hyperplasia positive expression than normalgastric mucosa (P <0.05); the Bitl positive expression in early cancer anddysplasia, normal gastric mucosa there is no significant difference (P>0.05). AIF the normal mucosa overexpression in advanced gastric cancerand gastric next rate are79.4%(27/34) and41.2%(14/34). Bit l proteinpositive signal is localized in the cytoplasm, pale yellow-yellow, theedge of the cancer nests the connective tissue positive, well-differentiatedcancer Bitl expression positive, poorly differentiated cancer cells negative.Bitl positive rate in advanced gastric cancer and adjacent normal gastricmucosa are35.3%(12/34) and38.2%(13/34). The analysis shows thatoverexpression of AIF in advanced gastric higher than that of normal gastric mucosa (P <0.05), and the positive expression of Bitl two is nosignificant difference (P>0.05). The AIF gene protein expression levelwith clinical stage and patients’ gender no significant correlation ingastric cancer tissues (P>0.05), Bitl gene protein expression levelscorrelated with sex (P>0.05), there is no obvious correlation but Bitlexpression significant differences in the different clinical staging ofgastric cancer, significantly higher than that in advanced cancer (P <0.05)in early cancer. AIF and Bit1protein are medium relatedness (r=0.395).Conclusions AIF over expression in gastric carcinoma than normalgastric mucosa, the AIF protein gene expression levels with clinicalstaging of patients with gastric cancer and no significant gender-related,and the expression prompted AIF over expression abnormalities may beassociated with the occurrence of gastric cancer related to thedevelopment. Higher than that of normal gastric mucosa Bitl positiveexpression in atypical hyperplasia tissues, the positive expression inadvanced gastric cancer and normal gastric mucosa is no significantdifference in protein expression level of Bitl genes and gender is nosignificant correlation. In expression of Bit l in different gastric clinicalstage difference significant expression in early cancer than in advancedcancer. The expression prompted Bitl abnormalities may be an early eventin gastric carcinogenesis. |
