| Objective:Pre-eclampsia is a multifactorial and polygenic disease, current research focuson the regulation of miRNA toward target gene post-transcription level.This studyaims to detect and analyze the differentially expressed miRNAs between sPE andnormal pregnancy placenta tissue, exploring the role of miRNA in sPE.Methods:1.Placenta tissues were obtained from eleven pregnancies complicated by sixsPE and five normal pregnancy women at the time of caesarean section who werehospitalized in Department of Gynecology and Obsterics of jiangxi province fromOctober1st,2011to April30st,2012.2.The total RNA was extractd with Trizol method whose concentration andpurity was measured by spectrophotometer.3.The sample RNA was marked with the miRCURYTMArray Power LabelingKit,then hybirdized with the7thgeneration of miRCURY LNATMmiRNA chip(including3100caputer probes,covering all human,mouse and rat microRNAsannotated in miRBase18.0,as well as all viral microRNAs related to thesespecies).The hybirdization results were scanned by Axon GenPix4000B chipscanner,analyzed by GenPix pro V6.0software.MEV software was used to dodifferentially expressed screening(with P value<0.05and Fold Change value≥1.5or≤0.5) and do cluter analysis.Results:1. Sixty miRNAs were differentially expressed between sPE and normalplacenta issues,and all of them were upregulated in placentas with sPE.2. miR-451has confirmed that differentially expressed in sPE placenta tissue, inaddition, this study found some abnormal expression of miRNA such as miR-767andmiR-203and miR-25, miR-218, etc.Conclusions:MicroRNA expresion profiles microrray is an effective high-flux method to detect differential miRNAs expression in sPE.The different miRNAs detected in themiRNA microarray analysis may be the new and characteristic moleular markers ofthe sPE,and could help us get better understanding of the miRNAs’ function insPE,and go further in searching for related biomolecular markers of clinical diagnosisand treatment in sPE. |
