Observation and analysis of differentiallyexpressed in retinal tissue and brain tissue of PrP~cmice18months of age.It provideexperimental basis for the relationship of PrP~c, Alzheimer’s disease (AD) and visuallesions.Methods:choose the healthy ICR mice at2weeks of age,9months age, the age of18months, the monthly age20, male and female. Fully expose eye and cut off the opticnerve transection of the posterior pole of eyeball, using microinstrument gentlequickly out the retina, retinal into EP tube,-80°cryopreservation. Using reversetranscription polymerase chain reaction, RT-PCR) expression of PrP~cgene wasdetected in retinal tissue; Western blotting (Western Blot, WB) PrP~cproteinexpression were detected in retinal tissue, The above two ways were used to detectthe expression of β-actin as control factor. Analysis of differentially expressed in therelationship between expression levels of PrP~c in the mouse retina with months and18months of age, the mouse retina and brain tissue of PrP~c.Results:1, The age of mouse tissue samples reference factor beta-actin is stable, not withthe age changing, the Institute confirmed by RT-PCR and Western Blot technology isstable and reliable, no obvious errors;2, At the age of2weeks,11months,18months of PrP~cgene in mice retinamonths expression level increased with age, the age of PrP~cexpression levels weresignificantly different (P<0.01); the expression of PrP~cprotein in mouse retina of each age level also along with the age increased, compared with significant differencesamong the groups (P<0.01);3, The PrP~cexpression level of18month old mice brain tissues is higher thanthe level of the same group of retinal expression, the difference was statisticallysignificant (P<0.01).Conclusions:To explore the expression of PrP~cgene in normal mouse retina, and theexpression level increased with increasing months of mice, the expression intensitybut weak in brain tissue and PrP~c. |
