Objective:Graft skin or artificial skin is needed for the repair of large skin woundrepairing. The study aims to R&D a biodegradable nanofiber membrane containingbioactive factors which would repair skin wound effectively, thus meeting themarket demand.Contents and Methods:1.The PCL fiber membrane, PCL/GE fiber membrane and PCL/GE/BFmembrane were made by high voltage Electrospinning and electrostatic spraytechnology accordingly. The physical-chemical properties of these fiber membraneswere studied by weight method, SEM, ATR-FTIR, Static contact Angle (SCA),degradation test, respectively.2.The biocompatibility and wound repair effects were studied by cytologyexperiment and animal experiment.Results:1.Characterization tests showed the porosity of PCL, PCL/GE, PCL/GE/BFmembranes from high to low is: PCL film> PCL/GE/BF> PCL/GE film, the densityof the menbranes from high to low is: PCL/GE film> PCL/GE/BF> PCL film, all themembranes have low density and high porosity, and the surface of fiber is smooth.2.The in vitro degradation experiment showed that the gelatin in fiber could notonly increase the PCL hydrophilic, but also accelerate the process of fiberdegradation.3.Cell-scaffold co-culture test showed the PCL/GE/BF, PCL/GE fiber membranehave good characteristics of cells adhesion and cell compatibility. Cell proliferationof PCL/GE/BF group was the most apparent; however, the effects of PCL group waslower than that of the two others.4.In vivo experiments and pathology slices from New Zealand rabbits woundmodels treated with various membrane showed PCL/GE/BF was beneficial to the early formation of the wound capillary, it could promote the wound repair obviouslyand had good protective effect for the wound, meanwhile, the protection effect of thePCL/GE membrane is better than that of the PCL membrane.Conclusion:As a new type of biodegradable nanometer scaffold with biological activity, thePCL/GE/BF has good biocompatibility, and it has a huge advantage in woundrepairing.
