Objective:To observe the effect of sildenafil in the treatment of persistent pulmonary hypertension (PPHN) of the newborn.Methods:Forty three neonates with PPHN were recruited from2012.10to2013.10in NICU of Zaozhuang Maternal and child Health Care Hospital. Among43newborns with PPHN,20cases were treated with the routine treatment only(group A), and23cases were treated with sildenafil additionally(group B). Several indexes including clinic outcomes, blood-gas analysis, radiologic and echocardiographic findings were observed.Results:There were more recovered cases in group B(65.22%)than in group A(40%)(P<0.05). Serial blood-gas analysis showed that cases in group B had better oxygenation indexes including PaO2, PaCO2, and OI than those in group A(P<0.05). PAP (pulmonary artery pressure)notably decreased in group B than in group A(P<0.05). The average hospitalization expenses and the average hospitalization time had no significant difference (P>0.05) in two groups. The time of using respirator in conventional treatment group was8.53±11.35days on average, while time with sildenafil treatment group was4.64±7.28days on average, and it had significant difference (P<0.05).Conclusion:1.PPHN treated with sildenafil has good effect compared with routine treatment only.;2.Sildenafil can be used to shorten the time of mechanical ventilation for neonates with PPHN. 3.Sildenafil can decrease PAP and improve oxygenation index; Sildenafil is an effective and safe drug in treating PPHN.4.Adverse reactions were not seen during therapy with sildenafil in all patients.
