Research objectives:The objective of this study is to demonstrate whether c-maf is expressed in multiple myeloma cell line of RPMI8226.Further studies were to investigate the effect of c-maf gene on the proliferation,invasion activity and cell cycle of RPMI8226cells.Research method:Lipofectin Reagent was used to transfect C-maf siRNA into multiple myeloma cell of RPMI8226.The mRNA expression level of c-maf was detected by RT-PCR.Cell growth curve was measured by MTT assay.Transwell chamber test was used to measure MM cell invasion activity.Results:1. By the fluoroscope we could easily see green fluorescence sending out by transfected MM cells.2. The mRNA expression of the c-maf gene was detected by RT-PCR in transfected cells. Data analysis was used to compare gene expression inhibition ratio of different groups.The result showed that the ratio of c-maf siRNA transfected group is (60.38±7.36)%,it is significant higher than other two group (P<0.05)3.For transwell chamber test,after RPMI8226cells had been transfected with c-maf siRNA,the average number of RPMI8226cells which migrated through the matrigel and filter from the upper chamber to the lower chamber in the c-maf transfected group,the NC-siRNA group and the Mock siRNA group was35.66±5.09、57.01±10.22、60.33±9.63respectively.The difference in the cell number between c-maf transfected group and the other two groups were statistically significant (P<0.01)4.MTT test was used to investigate cell survival rate of every group cell,after be transfected for24,48,72,96,120hr.Result showed when cells transfected for24hr,the difference is appeared,and the significant difference was occurred at48hr.The OD490values of the c-maf transfected group were0.26±0.08, it is significant lower than other two group (P<0.05)Conclusion:1.C-maf gene had a high expression in multiple myeloma cell lines of RPMI8226,we can effectively silence its overexpression by c-maf siRNA.2. C-maf siRNA can remarkably inhibit proliferation and invasion of multiple myeloma cell lines of RPMI8226.3. C-maf gene may be used as biological therapy for multiple myeloma target genes.
