Senile disease (ie, Alzheimer’s disease, AD) is one kind of brain dysfunction due to brain organic impairment and classified as degenerative nerve system diseases. The disease is characteristic for etiology, pathology and clinical manifestations. Etiologically, it was considered the kidney deficiency, phlegm accumulation, blood stasis as the basic pathogeneses in TCM, further more, mood and the specific physical factors of senile population can not be ignored. It is believed that the disease progression caused by multiple factor and complicated, but the specific pathogeneses was not entirely clear, the combined effect of genetic changes with environmental factors be possible. Pathologically, three main characteristics of senile plaques (SP), neurofibrillary tangles (NFT) and neuron loss were formed with the reason of β-amyloid protein disposition. From the clinical manifestations point of view, the incidence is cryptic with the obvious symptoms of progressive dementia, cognitive impairment accompanied by behavioral and psychological abnormalities. The chronic inflammatory reaction goes thoroughly in the onset and ongoing of the disease, and various glial cells and inflammatory factors involved. The inflammatory reaction and inflammatory factors was considered as the pathology nature of "yang-heat" in TCM. On the basis of referring literatures and combined with personal study understanding, it is believed that the yang-heat may be and important causative factor as well as pathological product in the development of the disease.During the study, on the basis of rat sub-acute aging by long term intra-abdominal injection of D-gal, the AD rats model were replicated by one-time intra-cerebral injection of Aβ1-42and Ibo mixture with the Huperzine A tablets as the control group. Observe the effect of "gailiang sanjiasan" which formulated principally by "tonifying kidney, activating blood circulation and resolving phlegm" on the disease indexed by behavioral indicators and several inflammatory factors. Result:"gailiang sanjiasan" can improve learning and memory, which may be achieved by precursor gene expression inhibition of P-amyloid protein to reduce the intra-cerebral factors IL-1α IL-1β、IL-6、NF-κB and the mitogen-activated protein kinase (MAPK) expression so as to reduce intra-cerebral inflammatory reaction, and the reduction of Aβ disposition finally achieve the goal of postponing the AD development.
