Association Between IL-1, IL-18and IL-33Gene Polymorphisms And Late-onset Alzheimer’s Disease

Objective:To examine whether the single nucleotide polymorphisms in interleukin(IL)-la gene(rs1800587), IL-1βgene(rs1143627), IL-18gene(rs187238) and IL-33gene(rs11792633) are associated with late-onset Alzheimer’s disease in Hunan Han Chinese population.Methods:We used a case-control study involving201LOAD patients and257age-matched and sex-matched controls from healthy subjects. The gene polymorphisms of rs1800587in IL-la, rs1143627in IL-1β, rs187238in IL-18and rs11792633in IL-33were detected by PCR amplification and Matrix-Assisted Laser Desorption/Ionization Time of Flight Mass Spectrometry(MALDI-TOF-MS), followed by analysis on the differences in genotype and allele frequencies between the AD group and the control group and association between the four SNPs and LOAD.Results:1. No significant differences were found in genotype frequencies of rs1800587between the two groups(P>0.05), but significant differences were found in the CC and (CT+TT) genotype and C, T allele frequencies between the two groups(.P<0.05), T allele of rs1800587was associated with a significantly increased risk of LOAD(OR=1.592,95%CI=1.037-2.443).2. No significant differences were found in genotype and allele frequencies of rs1143627or rs187238between the two groups(P>0.05).3. Significant differences in genotype and allele frequencies of rs11792633were found between the two groups(P<0.05), T allele of rs11792633was associated with a significantly lower risk of LOAD(OR=0.648,95%CI=0.498-0.843).4. Education, widowed, total cholesterol, IL-1α gene(rs1800587) and IL-33gene(rs11792633) were correlated with LOAD significantly, based on multivariate unconditional Logistic regression analysis.Conclusions:1. Our results indicate that the gene polymorphisms of rs1800587in IL-1α may contribute to the susceptibility to LOAD in Hunan Han Chinese popolation, the T allele was a risk factor of LOAD.2. The gene polymorphisms of rs1143627in IL-1β and rsl87238in IL-18do not appear to influence the susceptibility to LOAD in Hunan Han Chinese population.3. The gene polymorphisms of rs11792633in IL-33may contribute to the susceptibility to LOAD in Hunan Han Chinese population, the T allele was a protective factor of LOAD.
