In the full text, the curcumin in several of curcuma plants was extracted, separated anddetermined, at the same time the volatile components were analyzed.In the extraction, reflux and ultrasonic method are used in the single factor experimentsand the orthogonal test. The optimum extraction conditions of ultrasonic method were that theextraction reagent was90%ethanol as extraction reagent, solid-liquid ratio was1:10,ultrasonic temperature at50℃and ultrasonic time was50min, extracted three times. Theoptimum extraction conditions by of reflux method were that the extraction reagent was100%ethanol, solid-liquid ratio was1:10, reflux temperature at80℃and extraction time was60min, extracting extracted twice.In the purification, reflux with methylene chloride, extraction with sodium carbonatesolution and thin layer chromatography (TLC) were carried out.The optimum separationconditions with methylene chloride were that reflux time was2h, reflux temperature at41℃,and solid-liquid ratio was1:13, refluxed twice; the optimum separation conditions withsodium carbonate solution were that extraction solvent was1.5%sodium carbonate solution,extraction time was15min, extraction solution:extraction agent ratio1:1; The developingagent in thin layer chromatography was chloroform, methanol, formic acid=96:3:1(V: V:V).In the quantitation, HPLC was used for analysis of the concentration of curcumin,demethoxycurcumin and bisdemethoxycurcumin. In this study we chose the best conditionsby HPLC with Kromasil C18column as stationary phase,2%acetic acid solution: acetonitrile(60:40, V: V) as mobile phase; the flow rate was1.0mL/min; column temperature at25℃.The result was that in turmeric the concentration was the highest, in radices curcumae wasthe second and in rhizoma zedoariae was the lowest.In the same sample, the concentration ofcurcumin was the highest, which was much higher than demethoxycurcumin,bisdemethoxcurcumin, and the concentration of demethoxycurcumin andbisdemethoxycurcumin was or less the same.The research on volatile oil in several kinds of curcuma plants samples was carried outby solid phase microextraction and steam distillation, GC-MS was used for separation andidentification of volatile components. By solid phase microextraction method,24componentsin turmeric,31components in radices curcumae and37components in rhizoma zedoariaewere detected. By steam distillation,28components in turmeric,26components in radicescurcumae and49components in rhizoma zedoariae were detected. Those two kinds ofmethods detected many common components in the determination, such as caryophyllene,eucalyptus. Also there are a lot of proper components such as cyclosativene andCyclopropa[d]naphthalene, detected by solid phase microextraction method in turmeric;cubebene and elemene detected by solid phase microextraction method in rhizoma zedoariae;isoborneol, large root myrcene and aristolochic ene detected by steam distillation in rhizomazedoariae.
