Objective:Througth establishing the model of rabbit ear hyperplastic scar under the actiong of TGF-β1,and then observe the pathological changes, detect the PPAR-βmRNA and protein expression, and The expression of collagenous fibre of Ⅰ and Ⅲ in the scars,Thus investigate the relationship between TGF-β1and PPAR-β in the process of scarring.Methods:1. Twelve New Zealand white rabbit were choosen and lidocaine infiltration that rabbit after anesthesia, in its ears ventral side were two oblong incision of the length of the side2*1cm, complete resection of full-thickness skin, and burning hot wire and the skin around the edge, the left(experimental side) wound were applied transformation growth factor beta-1, and the right side (control side) to physiological saline topical, change a medical prescription every three days with the same agentia, observe the wound healing and the scar hyperplasia, after7,21,30,60,90,120days for scar tissue formed, using HE staining to observe the pathology test results; Rabbit ears hyperplastic scars tissue in7d,21d,30d,60d,90d and120d were detected by the immunohistochemical, RT-PCR and Western Blot to observe the expression of PPAR-β mRNA and protein, and the expression of collagenous I and III.Result:1. HE staining pathology results:In experimental group the keratinized layers much thicker than those in control group, and the collagen proliferation in derma more obvious;2. The PCR results: provided fluorescence quantitative PCR analysis software BIO-RAD CFX Manager for Statistical analysis, the raw data obtained with the SPSS13.0to T test, P<0.05, suggesting that PPAR-βmRNA expression of rabbit ears hyperplastic scar tissue in experimental group were higher than those in control group;3. WB results:by ImageJ analysis software to read the optical density value, the data obtained with the SPSS13.0to T test, P<0.05, suggesting that PPAR-βprotein expression of rabbit ears hyperplastic scar tissue in experimental group were higher than those in control group;4. The immunohistochemical results:The expression of collagenous Ⅰ and Ⅲ in the experimental side are higher than controlConclusion:1. The pathological structure of rabbit ear hypertrophic scar is similar to the human hypertrophic scar.;2.External use of TGF-β1could upregulate the expression of PPAR-βin early stage of rabbit ear hypertrophic scar;3.PPAR-Pmay could upregulate the expression of Ⅰ and Ⅲ collagen fibers in rabbit ear hypertrophic scar.
