Objective Allergic rhinitis is one of common diseases in E.N.T. department, whose pathogenesis has not be totally understood yet. And the current steroids hormone therapy for allergic rhinitis is not working well. The newly-presented specific immunotherapy of AR holds a very bright perspective, but on the other hand, it has the disadvantages of huge workload to detect all the sensitinogen for a specifical individual and longer course of the treatment, and the AR tissue remodeling may protract the course of disease and make the therapy more complex. In this study, regulatory factors and regulating networks which are closely associated to the outbreak of perennial allergic rhinitis and specific immunotherapy are selected to lay the foundation for further research on AR tissue remodeling.Methods45perennial allergic rhinitis patients,22SIT-effective and20SIT-ineffective allergic rhinitis patients and20healthy controls were recruited for this study. Isobaric tags for relative and absolute quantitation followed by two-dimensional gel electrophoresis and mass spectrometry were used to identify proteins that were differentially expressed among the four groups. The proteins that were significantly differentially expressed were clustered using EXPANDER v5.1software. The regulatory network pathways were identified, and the proteins associated with specific immunotherapy treatment were put into these pathways using Metacore software.Results Based on the predetermined210differential proteins,10groups of regulatory pathways with the minimum p values are constructed by Metacore pathway software. And we found that besides the coagulation pathway and the immune regulation pathway, which function in AR, the cytoskeleton remoulding—the actin pathway in RHoGTP enzyme regulation has also been activated.Conclusion By corresponding bioinformatics analysis, it can be conjectured that the cytoskeleton pathway in RhoGTPases regulation has taken a part in the tissue remodeling of AR. And through the analysis of the cytoskeleton pathway in RhoGTPases regulation, it can be inferred that RhoA/Rock signal play a role in the tissue remodeling of AR, especially in the following three aspect:AR mucosal epithelial cell injury, microvascular remodeling in AR mucosa lamina proprias and the deposition of extracellular matrix.
