Objective In a recent study,periostin is upregulated in the airway epithelia of patients with bronchial asthma and considered to be involved in airway remodeling under pathological station.Our study aimed to examine the expression of periostin in a mice model and evaluate its role in the process of inflammatory and nasal remodeling associated with allergic rhinitis.Methods Forty BALB/C mice were divided into four groups according to completely random design as following,group A(NS),group B(OVA),group C(low-dose of budesonide),group D(high-dose of budesonide).There were ten mice in each group.Female mice(group B,C,D)were sensitized by intraperitoneal injection of 25μg Ovalbumin(OVA;Sigma,USA)and 2mg aluminum hydroxide(Thermo,USA)dissolved in 50μl NS on days 0,7,14.The control group A were injected with commensurable NS.All animals expect group A were daily intranasal challenge with 10μl OVA in each nostril from day 22 to day 29.Similarly,group A were challenged by same doses of NS.We measured and recorded the number of sneezes,nasal rubbing,as well as the condition of runny nose for 30 minutes after the last challenge at the fourth weekend.Prolonged inflammation was induced by subsequent nasal exposure to 10μl of OVA twice a week for 12 weeks.Meanwhile,group C,D were treated daily with intranasal budesonide following the doses of 3.0μg/20 g,15μg/20 g respectively for 12 weeks.The number of sneezes,nasal rubbing that occurred were counted for 15 minutes after the last treatment and then sacrificed after anesthetic.The two groups,C and D,received different doses of budesonide(3.0μg/20 g,15.0μg/20g)in order to establish inordinately murine model of allergic rhinitis.We evaluated nasal symptoms at the end of this experiment.In addition,we measured the levels of OVA-s Ig E and periostin in serum and IL-4,IL-13 in nasal lavage fluid with ELISA,as well as the expression of periostin in nasal tissue by immunohistochemistry.Statistical analysis was performed with 17.0 SPSS.A P-value of less than 0.05 was considered to be statistical significance.Results(1)The difference of the concentration of periostin in serum in group A-D was significant statistically(P<0.05).The concentration of OVA-s Ig E in serum in group A-D were.The differences among these four groups were significant statistically(P<0.05).The concentration of IL-4 in nasal lavage fluid in group C and D were significantly increased than in group B(P<0.05).However,there were no significant differences between group D and A when compared with each other.The concentration of IL-13 in nasal lavage fluid in group C and D significantly increased than in group B(P < 0.05).There were no significant differences between group D and A when compared with each other.The numbers of eosinophils in group C and D significantly increased than in group B(P<0.05).There were no significant differences between group D and A when compared with each other.(2)The differences of the diamet-ers of gland cells among these four groups were significant statistically(P<0.05).The differences of the number of goblet cells among these four groups were significant statistically(P<0.05).As for sub epithelial thickness,group C and D significantly increased than group B(P<0.05).No significant differences could be concerned between group D and A when compared with each other.Conclusions(1)Nasal remodeling can be induced by Ovalbumin challenge in AR mice.(2)Periostin is expressed in the nasal mucosa of murine with allergic rhinitis.(3)It may play an important role in subepithelial fibrosis and eosinophilic inflammation,affecting nasal mucosa remodeling in allergic rhinitis.
