Objective:This study is to explore the association between the polymorphism of ACE, KLK1, KNG1genes and cerebral infarction in Han of Changsha.Methods:Collecting the cerebral infarction patients as the cerebral infarction group who were in the department of Neurology, The Third Xiangya Hospital of Central South University from June2011to July2012. The control group was consisted of healthy people who examined in the Third Xiangya Hospital. All subjects were Han in Changsha area and given informed consent. We examined single-nucleotide polymorphisms (SNPs) of genes involved in the kinin metabolism pathway in attempting to examine whether SNPs would confer a significant susceptibility to cerebral infarction by MALDL-TOF-MS. The results of genotyping were analyzed by chi-square test and logistic regression, to explore the association between the polymorphism of ACE, KLK1, KNG1genes and cerebral infarction in Han of Changsha.Results:1. All these three SNPs distribution of gene ACE, KLK1, KNGl in the population are in line with Hardy-weinberg equilibrium.2. We complete an association analysis of distribution of genotype and allele frequencies by Chi-square test using the case control association tests. The results are as follows:①the genotype and frequency of allele in rs4343of ACE gene were statistically different between cerebral infarction patients and control subjects (p<0.05).②The genotype and allele frequency in rs5517of KLK1gene and rs710446of KNG1gene were found not different between cerebral infarction patients and control subjects (p>0.05).3. We verified the preliminary results by logistic regression, and corrected high risk factors such as BMI, blood glucose, blood pressure, HDL, LDL, TC, TG. The results are as follows:①the genotypes distributions in rs4343of ACE gene and rs5517of KLK1gene have no association with cerebral infarction (p>0.05);②the AA genotype in rs710446of KNG1gene was found different between cerebral infarction patients and control subjects (p<0.05), whose odds ratio was0.66, which means it was a protective factor of cerebral infarction.Conclusion:There was association between the polymorphisms of rs710446in KNG1gene and cerebral infarction in Han of Changsha. There were no association between the polymorphisms of rs5517in KLK1gene and rs4343in ACE gene and cerebral infarction in Han of Changsha.
