Objective:In this study we investigate the effects of DADS on human gastric cancer BGC-823on cell proliferation and apoptosis, Aimed at analysis the possibility of consider DADS as effective antineoplastic.Methods:(1) The cell growth inhibition of BGC-823and HUVEC were measured by MTT assay.(2) Morphological changes of the two kinds of cells were observed by microscope.(3) Cell cycle distribution and apoptosis of BGC-823and HUVEC were analysed by FCM.(4) Western blot was applied to detect the expression of Bcl-2protein, Bak protein and P53protein of the two kinds of cells.Results:(1) MTT results have shown that DADS inhibite the proliferation of BGC-823cells in a dose-dependent manner, and in the concentration of62.5μM~125μM the inhibition of BGC-823cells is bigger than HUVEC cells.(2) After treated with DADS, some of BGC-823cells began to shrinking, rounding and so on, but HUVEC cells acted these changes only when DADS is500μM.(3) FCM showed that DADS can only reduce a small part of BGC-823cells and HUVEC cells to apoptosis, and the apoptosis ratio of BGC-823cells is higher than HUVEC cells. The cell cycle of both cells did not change obviously.(4) After treated with DADS, BGC-823cells acted as down-regulation of Bcl-2protein, and the expression of Bak and P53protein were not significantly changed. The expression of Bcl-2, Bak and P53of HUVEC cells were not significantly changed.Conclusion:DADS can inhibit the proliferation of BGC-823cells, but maybe cell cycle arrest and apoptosis are not the main reseans, when DADS is at the concentration of62.5μM~125μM the inhibition of BGC-823cells is higher than HUVEC cells.
