Objective:To investigate the relationship between adenosine concentration in renal allograft and allograft fibrosis.Methods:Wistar kidneys were orthotopically transplanted into SD rat recipients (Experiment group). SD to SD rat kidney transplantation was served as the control (control group). Both control and experimental group divide into1week,2weeks,3weeks,5weeks groups post-transplantation. Animals were sacrificed1-5weeks post-transplantation for histological and immunohistochemical detection. HPLC method was used for determination of renal allograft tissue adenosine concentration. HE staining to observe the graft tissue inflammation and histopathological changes. Masson’s trichrome staining to observe the collagen deposition in renal graft. Immunohistochemical method was used for determination of TGF-β1and a-SMA expression; Real-time PCR method was used for determination of fibrotic genes TGF-β1, pro-collagen Ⅰ (α1) mRNA expression; Real-time PCR and immunohistochemical method were used to detect the degree of renal fibrosis. So We can analyzed the correlation between ADO levels in graft tissue, the degree of acute rejection and renal graft fibrosis;Results:Compared with control group, the lever of adenosine increased significantly in experiment group which exsits acute rejection. The progression of allograft fibrosis is accelerated, which show that collagen deposition, the fibrotic genes TGF-beta1and Pro-collagen mRNA expression and TGF-betal and a-SMA protein expression was significantly increased.Conclusion:Our study suggests that Elevated adenosine has a high correlation with acute rejection and allograft fibrosis. Increasing of adenosine in allograft post-transplantation plays an important role in kidney fibrosis originate from acute rejection. Acute rejection leading to renal allograft fibrosis may via the adenosine signaling pathways.
