Objective:Discuss the correlation of sperm-originated miRNA and cleavage stage embryo quality by detecting the mir-21、mir-34c、 mir-140、mir-375expression in the sperm of IVF patients with different embryo qualityMethods:The remaining fresh semen specimens were collected from44male patients who were in the IVF cycle of the Xiangya Hospital Reproductive Center from September to December in2012, the expression of sperm microRNAs (mir-34c、mir-140, mir-21, mir-375) were detected by Real-time fluorescent quantitative PCR. The number of embryonic cells, uniformity of blastomere and fragments were observed on the second and third day, the embryo were divided into two groups after scoring according to embryo criteria, one group of which the average embryo score<8is regarded as the exprimental group, the other is the control group. Compared the general date and experimental date of the exprimental group and the control group, analyzed the correlation of the miRNA expression in sperm of the44cases and embryo score. Result:1.The experimental group showed a lower expression of mir-34c、 mir-140, mir-21, mir-375in sperm, there was significant difference compared with the control group (P<0.05).2.Retrieved follicles、metaphase Ⅱ stage ovocytes、fertilized oocytes, cleavage number、fertilization rate in the experimental group without significant difference compared with the control group (P>0.05); the cleavage rate on the second day in the experimental group is lower than that of the control group, which had statistical significance(P<0.05).3.There was a negative correlation between the expression level of mir-21in sperm and the ratio of fragment on the second day (r=-0.652,P<0.01) and the ratio of fragment on the third day (r=-0.707,P<0.05).The expression level of mir-21was positively correlated with the embryo score (r=0.809,P<0.01) and the blastomere quantity on the third day (r=0.743,P<0.01).4.There was a negative correlation between the expression level of mir-34c in sperm and the ratio of fragment on the second day (r=-0.644,P<0.01) and the ratio of fragment on the third day (r=-0.706,P<0.01);the expression level of mir-34c was positively correlated with the embryo score (r=0.874,P<0.01) and the blastomere quantity on the third day (r=0.715,P<0.01). 5.There was a negative correlation between the expression level of mir-140in sperm and the ratio of fragment on the second day (r=-0.612, P<0.01and the ratio of fragment on the third day (r=-0.604, P<0.01);the expression level of mir-34c was positively correlated with the embryo score (r=0.848,P<0.01) and the blastomere quantity on the third day ((r=0.632,P<0.01)6. There was a negative correlation between the expression level of mir-375in sperm and the ratio of fragment on the second day (r=-0.616, P<0.01) and the ratio of fragment on the third day (r=-0.651, P<0.01).The expression level of mir-375was positively correlated with the embryo score (r=0.809, P<0.01) and the blastomere quantity on the third day (r=0.724, P<0.01).Conclusion:The expression level of mir-21,mir-34c, mir-140, mir-375in sperm may have an important role as positive regulators in the development of Cleavage stage embryonic and thus influence embryonic quantity.
