Objective To study the effect of the extraction of Prunella Vulgaris. on anti-MDR-TB, and to make the preliminary exploration of how drug-resistant TB in the formulation of the clinical scheme has significant value for reference, facilitating modernization study of traditional Chinese drug.Methods (1) Sputum specimens from typical pneumoconiosis tuberculosis were collected, then Multidrug-resistant tuberculosis (MDR-TB) were cultured and identified; The drug-resistant gene of katG, rpoB and rpsL were amplified from MDR DNA, and the positive specimens of MDR-TB were detected by single strand cinformation polymorphism analysis (SSCP).(2) MDR-TB model in rats was induced by MDR-TB. Normal control group, model group and Prunella Vulgaris group were set up. The content of IFN-y, IL-4, IL-10and IL-12were examined by ELISA. RT-PCR was used to detect the mRNA levels of them.Results (1) The positive test results of Mycobacterium tuberculosis bacteria were acquired in104cases of194pneumoconiosis tuberculosis specimens, and the total detection rate was53.61%(104/194). The detection rate of MTB strains resistant to INH, RFP and SMU were76.92%(80/104),90.38%(94/104) and50.00%(52/104) through method of AST, and among these the MDR-TB strains were80(76.92%), including42strains resistant to3types of drugs simultaneously (40.38%),38strains resistant to2types of drugs simultaneously (36.54%), and24strains resistant to single drug (23.08%). The mutation rate of katG, rpoB, and rpsL was57.70%(60/104),65.38%(64/104),40.38%(42/104) respectively.30strains were detected combination mutations of the three genes (30/104,28.84%), and the compliance rate of detection was84.21%(32/38).12strains were detected single gene mutation (12/104,11.54%), and the compliance rate was50.00%(12/24). Among strains resistant to drugs, katG gene showed negative in20strains, rpoB gene showed negative in30strains, rpsL gene showed negative in10strains.(2) The results of ELISA showed that Prunella Vulgaris group resulted in significant in serum IFN-y, IL-12, IL-4and IL-10(P<0.05or P<0.01), the mRNA levels of them were significant (P<0.05or P<0.01).Conclusions From detecting the content changes of cytokines of IFN-y, IL-4, IL-10and IL-12closely related with immunity in tuberculosis by ELISA, and at the same time using RT-PCR to detect the gene express of mRNA in these, it is show that the extraction of Prunella Vulgaris can enhance the cellar immunological function in rats from up-regulation of the level of genetic transcription, accordingly it has a conspicuous anti-MDR-TB effect.
