Objective:To explore the effects and mechanisms of KAI1gene on proliferation andinvasion and metastasis of gastric carcinoma cells, to study the expression of KAI1ingastric cancer and their correlation with clinicopathologic features. And investigatethe expression of ARTN and VEGF, to explore the regulation of its expression and therelationship among KAI1and ARTN and VEGF and explore the clincial value.Methods:The KAIl gene was transferred into human gastric carcinoma cells by theliposome. The expression KAI1mRNA after transfection was measured by RT-PCRmethod. MTT methods examine the cell proliferation. The invasive and metasticability of gastric carcinoma cells were analyzed with transwell culture methods.The expression of KAI1protein and ARTN and VEGF protein of gastric cancertissues was examinated by S-P immunohistochemical methods. Then combining withthe features of clincial pathology, we analyzed the relationship between the theexpression of them and these factors, and explore its regulation by PEMS3.1.Results:The KAI1mRNA was up-regulated in the cells transfected by KAI1gene. Thetotal positive rate of KAI1protein was23.3%in gastric cancer.The ability of proliferand invasive and metastic of gastric carcinoma cells was down-regulated.Expressionof KAI1protein was related to the degree of differentiation, the invasion depth, lymphnode metastasis and the tumor clinical stage(P<0.05). And the expression of ARTNprotein was positively correlated with VEGF protein, both was negatively correlatedwith the expression of KAI1protein.Conclusion:The KAI1mRNA was up-regulated in the cells transfected by KAI1gene. Theability of prolifer and invasive and metastic of gastric carcinoma cells was down-regulated. It can help to evaluate the clinical stage and direct therapy. And playan important role in the occurence and development of gastric cancer, The expressionof ARTN protein in gastric cancer tissues is closely related to the tumor invasion andmetasis, was positively correlated with expression of VEGF protein, all wasnegatively correlated with the expression of KAI1protein, can be used as one of theclinical indicators of the evaluation of progression in gastric cancer.
