Preliminary Mechanisms Of Therapeutic Effect Of Hao Bie Yang Yin Ruan Jian Prescription On Rat Hepatic Fibrosis Induced By Bovine Serum Albumin

Objective:To study the effect and mechanisms of Hao Bie Yang Yin Ruan Jian prescription on immuno-injured hepatic fibrosis induced by bovine serum albumin in rat model. To provide experimental basis of hepatic fibrosis for clinical use of Hao Bie Yang Yin Ruan Jian prescription.Method:The model of immuno-injured hepatic fibrosis induced by bovine serum albumin was established in Wistar rats. Bovine serum albumin containing incomplete Freund’s adjuvant was used to immunize rats by the method of subcutaneous injection (except for normal group),5times, then through the double diffusion agar plate method to confirm whether or not BSA antibody was induced, and then by the rat tail vein to inject bovine serum albumin,2times a week,15times before model success. Rats were divided into6groups:model group, high dose of Hao Bie Yang Yin Ruan Jian prescription, middle dose of Hao Bie Yang Yin Ruan Jian prescription, low dose of Hao Bie Yang Yin Ruan Jian prescription, complex Biejia Ruangan tablet group and qiu shui xian jian group by the method of randomized block. At the end of modeling, drugs were given to the corresponding therapeutic groups, and then were continued at the same time for two months. Distilled water was given to the rats of normal and model groups according to the same method. Liver tissues and blood serum were obtained at the end of the experiments. Liver tissues were used for histological examination, pathological grading, measuring the content of collagen protein, a-SMA and apoptosis of stromal cells. Blood serums were used for measuring the index of hepatic function.Results:1. Histological examination and pathological grading:The degree of hepatic fibrosis in model group was significantly higher compared with normal group (P<0.01). The degree of that in every therapy groups was significantly decreased (P<0.01, P<0.05). And there was no significantly difference between therapy groups of Hao Bie Yang Yin Ruan Jian prescription and another therapy groups.2. The contents of collagen protein of hepatic tissue:The contents of collagen protein in model group was significantly higher compared with normal group (P<0.01). The high dose of Hao Bie Yang Yin Ruan Jian prescription and middle dose of Hao Bie Yang Yin Ruan Jian prescription were significantly lower than model group. And there was no significantly difference between them and another therapy groups.3. The contents of AST, ALT, ALB, total serum protein and ALB/GLB:The contents of ALT and AST in model group was significantly higher compared with normal group (P<0.05), every therapy groups (except for low dose of Hao Bie Yang Yin Ruan Jian prescription) was significantly lower than model group. There was no significantly difference between every therapy groups of Hao Bie Yang Yin Ruan Jian prescription and another therapy groups. The content of ALB and value of ALB/GLB in model group were significantly lower than the normal group(P<0.05), The contents of ALB in high dose of Hao Bie Yang Yin Ruan Jian prescription and complex Biejia Ruangan tablet group were significantly increased(P<0.05). The value of ALB/GLB in complex Biejia Ruangan tablet group was significantly increased, and the value of ALB/GLB in high dose of Hao Bie Yang Yin Ruan Jian prescription group had tendency of increase. There was no significantly difference between high dose of Hao Bie Yang Yin Ruan Jian prescription group and complex Biejia Ruangan tablet group.4.a-SMA:The degree of hepatic fibrosis in model group was significantly higher compared with normal group (P<0.01). The degree of hepatic fibrosis in every group of Hao Bie Yang Yin Ruan Jian prescription was significantly lower than the post-treatment model group (P<0.01, P<0.05).5. Apoptosis of HSC:The value of pretherapy model group was significantly higher than that of normal group (P<0.05). The value of middle dose of Hao Bie Yang Yin Ruan Jian prescription was significantly higher than that of post-treatment model group (P<0.05).Conclusion:1. The model of hepatic fibrosis of rat was successful.2. Hao Bie Yang Yin Ruan Jian prescription has a therapeutic effect on hepatic fibrosis induced by bovine serum albumin.3. The mechanism of its effects might be associated with control activation of HSC, acceleration apoptosis of HSC and decrease injuries of hepatic cells.
