ObjectiveVia the studay the relevant factors in the signaling pathway of theAngiogenesis mimicry in Malignant melanoma, and then discusses theinfluence and mechanism of the RCG on malignant melanoma.MethodsB16melanoma cells were cultivated in vitro,then inoculated in right oxterin the C57BL/6mice.the model of mouse with Malignant Melanoma wasmade.the experimental mice were randomLy divided into normal salinegroup,RCG group at varied concentrations and CTX group.each groupconsisted of eight mice.Hight dosage group(700mg/kg),middle dosagegroup(300mg/kg),low dosage group(100mg/kg) and CTX group(60mg/kg)were injected with RCG and CTX group once a week.the grouth of tumor wasobserved and the inhibitory rate of tumor was measured.the protein expressionof EphA2, PI3K, LN-5γ2in tumor were determined with western blot,the geneexpression of EphA2, PI3K, LN-5γ2were detected with RT-PCR.ResultsThere is a founding that RCG has certain inhibitory effect on melanomavia giving RCG to the model of mouse melanoma(P<0.01),The proteinexpression level of EphA2, PI3K, LN-5γ2in RCG different groups were lowerthan saline group(P<0.01),the gene expression of EphA2, PI3K, LN-5γ2inRCG different groups were lower than saline group(P<0.01),and with the increase of RCG’s concentration, the protein expression level is lower.ConclusionRCG could effectively inhibit the growth of Malignant Melanoma inC57BL/6mouse, which was probably the result that RCG inhibit the proteinexpression of EphA2, PI3K, LN-5γ2in signaling pathway of the Angiogenesismimicry in Malignant melanoma.
