Objective:To explore the effect of the Jianpi Quyu Decoction intervene the VEGF/VEGFR2signal transduction pathway of retinal pigment epithelium-Bruch’s membrane-choriocapillaris complex of the dyslipidemia apolipoprotein E-deficient (ApoE-/-) mice animal models.Methods:2-month old ApoE-/-mice were randomly divided into the model group fed with high fat chaw and the control group fed with the normal chaw.4months later after determine AMD animal model have established, the model animals were randomly divided into the low dose group and the medium dose group and the high dose group of Jianpi Quyu Decoction and the controlled group, the four group are lavaged for1month.Observed all of groups the body mass, hemorheology, blood lipids change; the RBCC morphology was observed by optical microscope and the ultrastructural was observed by the transmission electron microscopy; the immunohistochemical method detection RBCC vascular endothelial growth factor (VEGF) and its R2receptor (VEGFR2) expression.and the retinal VEGF were analysed by Western-blot.Results:1.The body quality, hemorheology, blood lipid:Compared to the controlled group, the body quality of the medium dose group significantly reduced(P<0.05),the triacylglycerol,total cholesterol.Low Density Lipoprotein,plasma viscosity,Hematocrit of the medium dose group are significantly decreased (P<0.05)2. The retinal pigment epithelium (RPE) and Bruch’s membrane thickness: Compared to the controlled group, the RPE thinkness of the low dose group, medium dose group, high dose group is no significant changed (P>0.05). Compared to the controlled group, the Bruch’s membrane of the low dose group, the medium dose group and the high dose group are significantly thinner(P<0.05). 3. VEGF and VEGFR2in RBCC and retinal VEGF protein in the relative expression quantity:Compared to the controlled group, the expression quantity of the low dose group and the medium dose group are significantly decreased (P<0.05), compared to the low dose and the high dose group, the medium dose group are statistically decreased (P<0.05).Conclusion:1.7-month-old experimental dyslipidemia ApoE-/-mice pathological and physiological changes are similar to early AMD.2. To some extent, medium dose group and high dose group of Jianpi Quyu Decoction can improve the RBCC pathological damage of dyslipidemia ApoE-/-mice.3. The VEGF/VEGFR2signal transduction pathway may regulated by the Jianpi Quyu Decoction through declining the expression of VEGF, VEGFR2in RBCC.
