Dyslipidemia Lack Of Apoe Gene In Mice Rpe Bruch Membrane - Choroid Blood Capillary Complex Experimental Research

Object:To Observe the change of retinal pigment epithelium-Bruch’s membrane-choriocapillaris complex (RBCC) in apolipoprotein E-deficient (ApoE-/-) mice that effected by dyslipidemia, selected the experimental animals of age-related macular degeneration (AMD). Methods:Twenty-four2-month-old ApoE-/-mice and248-month-old C57BL mice were randomly divided into group of high fat chow and group of normal chow,12mice each group.4months later, four groups were executed and checked the test index, all of serum lipid and hemorheology index, RBCC of the mice eyeballs were observed under light microscope and transmission electron microscope by immunohistochemistry staining and Masson staining and retina by Western blot. The results were analyzed according to the SPSS for windows17.0software.Results:The blood biochemistry index and weight in the group of6-month-old ApoE-/-mice high fat chow were much higher than that of others; them in the group of6-month-old C57BL mice high fat chow were much higher than the group of6-month-old C57BL mice normal chow.The RPE thickness in the group of6-month-old ApoE-/-mice high fat chow were much thinner than that of others;the RPE thickness in the group of6-month-old C57BL mice high fat chow and the group of6-month-old C57BL mice normal chow has no difference. The Bruch membrane thickness in the group of6-month-old ApoE-/-mice high fat chow were much thinner than that of others; the Bruch membrane thickness in the group of6-month-old C57BL mice high fat chow were much higher than the group of6-month-old C57BL mice normal chow. VEGF and VEGFR2dyeing area and integral optical density by immunohistochemistry staining in the group of6-month-old ApoE-/-mice high fat chow was higher than that of others;VEGF in the group of6-month-old C57BL mice high fat chow were much higher than the group of6-month-old C57BL mice normal chow;but VEGFR2is not statistically significant between the group of6-month-old C57BL mice high fat chow and the group of6-month-old C57BL mice normal chow. Protein expression in retina by Western blot in the group of6-month-old ApoE-/-mice high fat chow was higher than that of others but there is not statistically significant between the group of6-month-old C57BL mice high fat chow and the group of6-month-old C57BL mice normal chow.Conclusion:1. The pathology change of RBCC in the group of6-month-old ApoE-/-mice high fat chow and the group of6-month-old C57BL mice high fat chow were similar to the change of AMD.2. In the group of6-month-old ApoE-/-mice high fat chow the change of VEGF and VEGFR2dyeing area and integral optical density by immunohistochemistry staining and protein expression in retina by Western blot were similar to the change of AMD.3. Compared the change of the group of ApoE-/-mice high fat chow and the group of C57BL mice high fat chow, the latter and age-related macular degeneration changes more similar, so ApoE-/-mice as age-related macular degeneration experimental animals.
