Object:The previous reseach hinted that sinomenine can inhibit the expression of downstream signal molecules and the transduction of signal which are belong to pattern recognition receptor,but the mechanism has no in-depth study. The subject posits that sinomenine cured rheumatoid arthritis(RA) by impact on the pattern recognition receptor. Through the study of organic tissue and inflammatory mediator to research for the relationship between pattern recognition receptor and the treatmen of RA when use sinomenine, inorder to provide laboratory evidence for post-marketing reevaluation of sinomenineMethods:Randomly we divide50male rats into five groups,then we build chicken type II collagen induced arthritis model in rats(CIA). We choosed some index to compare the differences between sinomenine’s high,medium,low treatment group and the model group. We measured the weight and swelling of rats periodically;we make pathorphological observation of rat’s synovial tissue of ankle joint(HE Staining);We detect the expression rate of TLR-2,NF-κB and TNF-α、IL-1which are come form rat’s synovial tissue of ankle joint.Finally we analyze all date with statistical software of spssResult:1、The weight growth of Rheumatoid arthritis rat model is slowed compared with control group,Sinomenine can effectively restrain this phenomenon.2、The Sinomenine can effectively reduce ankle swelling in rats.3、Sinomenine can reduce the expression levels of rat TLR-2. It can inhibit the CIA index of NF-κB,TNF-α.IL-1.it can also inhibit injury of synovial and ossa articularia.4、With the increase of the dose,the degree of the swelling becomes lower,the damage of synovial and ossa articularia becomes more slight,also the expression rate of the correlative index becomes more minor.5、Compared with The weight of medium,low Sinomenine group the high-dose group is more light.Conclusion:Sinomenine can inhibit the expression of TLR-2of pattern recognition receptor of APC cells,then the activation of NF-κ in the downstream is restrained.The NF-κB signalChannel is suppressed,it will block the activation of the APC which to make the production and secretion of IL-1,INF-α and other cytokines become less, it will also inhibit the activation of the synovial which can inhibit and delay the inflammation and destruction of the bone.All these can achieve the goal of anti-rheumatoid arthritis.
