In this study, a high performance liquid chromatography (HPLC) method wasused for the chromatographic fingerprint analysis of gardenia jasminoides, six batchesof Semen Sojae Praeparatum, soybean and Zhi Zi Chi Tang. The research indicatedthere was a significant variability in different batches of Semen Sojae Praeparatum,some major chromatographic peaks were absent in some batches of Semen SojaePraeparatum. An ASE method was adopted to extract the compounds in gardeniajasminoides and Semen Sojae Praeparatum after an investigation of three scavengemethods. A reliable procedure has been developed for identification of chemicalconstituents from gardenia jasminoides,Semen Sojae Praeparatum and Zhi Zi Chi Tangrespectively by high-performance liquid chromatography coupled with photodiodearray, electrospray ionization-tandem mass spectrometry(HPLC-DAD-ESI-MS/MS).19compounds from Semen Sojae Praeparatum,22compounds from gardeniajasminoides and25compounds from Zhi Zi Chi Tang were tentatively identified basedon the base peak ion (BPI) chromatograms and absorption spectra with reported valuesin the literature respectively.The comparative analysis of ingredients from gardeniajasminoides,Semen Sojae Praeparatum and Zhi Zi Chi Tang was established by usingelectrospray ionization-tandem mass as detector, The extract of Zhi Zi Chi Tang wasinitially separated and purified using D101macroporous resin by gradient elution ofdiffentent proportion ethanol meanwhile combine with HPLC. A confirmatory methodhas been developed for preparative separation and purification of active compoundsfrom Zhi Zi Chi Tang by macroporous resin and HSCCC, and7active monomers fromZhi Zi Chi were obtained by HSCCC for the first time.
